苍术麸炒前后对脾虚模型大鼠胃肠动力学的影响 被引量:12

E ffect of Raw or Bran-fried Rhizoma Atractylodis on Gastrointestinal Motility of Experimental Spleen Deficiency Rats

摘要 目的探讨苍术麸炒前后对脾虚大鼠胃肠动力学的影响,并探讨其可能的作用机制。方法将雄性SD大鼠随机分为正常组、模型组、生苍术组(10.0g·kg-1)、麸炒苍术组(10.0g·kg-1)、多潘立酮组(5.0mg·kg-1),共5组,每组10只。除正常组外,其余各组喂饲小承气汤煎剂加饥饱失常建立脾虚证大鼠模型,建模15d。建模结束后,给予相应药物灌胃,正常组及模型组给予等量生理盐水,每天1次,连续10d。10d后所有大鼠均用炭末灌胃法行大鼠胃内残留率、小肠推进比的测定,腹主动脉采血用放射免疫法检测血浆胃动素(MTL)、P物质(SP)、生长抑素(SS)含量,比色法测定大鼠空肠上皮细胞三磷酸腺苷(ATP)的含量,免疫组化法测定大鼠胃窦平滑肌细胞内肌球蛋白轻链激酶(MLCK)及结肠组织中干细胞因子(c-kit)表达量。结果与模型组比较,正常组、生苍术组、麸炒苍术组、多潘立酮组的胃内残留率明显下降、小肠推进比明显升高,而大鼠血浆MTL、SP和SS含量均不同程度升高,差异具有统计学意义(P<0.05,P<0.01)。与模型组比较,生苍术组、麸炒苍术组、多潘立酮组大鼠空肠上皮细胞ATP的含量及胃窦平滑肌细胞内MLCK和结肠组织中c-kit的表达量不同程度升高,差异均有统计学意义(P<0.05,P<0.01);与生苍术组比较,麸炒苍术组上述作用更加明显,差异均有统计学意义(P<0.05,P<0.01)。结论苍术提取物可从不同方面整体改善由于脾虚而导致的大鼠胃肠功能障碍,对因脾虚而导致的胃肠功能紊乱有较好的调节和治疗作用,且麸炒苍术的上述作用优于生苍术。 Objective To observe the effect of raw or bran-fried Rhizoma Atractylodis(RA) on gastrointestinal motility of rats with experimental spleen deficiency syndrome,and to explore the possible mechanism.Methods Fifty male SD rats were evenly randomized into normal group,model group,raw RA(10.0 g·kg^-1) group,bran-fried RA(10.0 g·kg^-1)group,and domperidone(5.0 mg·kg^-1)group.Except for the normal group,the rats in other groups were fed with Xiaochengqi Decoction plus irregular diet to induce spleen deficiency syndrome,and the modeling time covered 15 d.After the establishment of model,the medication groups were given the corresponding medicine,and the normal group and model group were given same volume of saline once a day for 10 days.After treatment,gastric emptying rate and intestinal propulsion proportion were evaluated after gastric gavage of Carbo Medicinalis,blood was sampled from abdominal aortic to detect rat plasma motilin(MTL),substance P(SP)and somatostatin(SS) with radioimmunity.Colorimetric assay was used for the determination of ATP content of rat jejunum epithelial cells,and immunohistochemical method was used to detect the expression of myosin light chain kinase(MLCK) in rat gastric antrum smooth muscle cells and c-kit in colonic tissues.Results Compared with the model group,the gastric emptying rate was decreased and intestinal propulsion proportion was increased significantly in the normal group,raw RA group,bran-fried RA group,and domperidone group,while the plasma MTL,SP and SS contents were increased to different degrees,and ATP content of jejunum epithelial cells,MLCK expression of rat gastric antrum smooth muscle cells and c-kit expression of colonic tissues were also increased,the differences being statistically significant(P〈0.05,P〈0.01).Compared with raw RA group,bran-fried RA group had more obvious effect,and the differences between the two groups were statistically significant(P〈0.05,P〈0.01).Conclusion RA extract can exerts an overall improvement of rat gastrointestinal dysfunction induced by spleen deficiency,and the effect of bran-fried RA is superior to that of raw RA group.
