摘要
目的研究破格救心汤中生附子剂量及煎煮时间对双酯型乌头碱的溶出量的影响。方法采用高效液相色谱法(HPLC),检测破格救心汤临床煎煮过程中双酯型乌头碱溶出量的动态变化,明确双酯型乌头碱溶出量与生附子剂量和煎煮时间的关系。结果以甲醇(0.1%三乙胺)-水(70∶30)为流动相,流速1.0 m L·min-1,柱温35℃,检测波长235 nm条件下进行测定。结果显示,复方中3种双酯型乌头碱溶出比例变化不大,其溶出量符合新乌头碱>次乌头碱>乌头碱这一趋势,其溶出规律为先增后减,且2 h达到最大值。结论该方法学考察符合定量要求,结果准确,可以解释破格救心汤水煎液中双酯型乌头碱溶出量的动态变化规律。
Objective To research the relationship of the dissolution content of diester diterpenoids alkaloids in Poge Jiuxin decoction with Radix Aconiti dosage and decocting time.Methods High performance liquid chromatography(HPLC)was used to study the dynamic variation of diester diterpenoids alkaloids in Poge Jiuxin decoction,and the relationship of diester diterpenoids alkaloids content with Radix Aconiti dosage and decocting time was explored.Results The detecting condition was as follows : the mobile phase system was composed of methanol(0.1 %triethylamine)-H2O(volume fraction being=70∶30),the flow rate was 1.0 m L·min^-1,the column temperature was35 ℃ and the detection wavelength was 235 nm.The results showed that total contents of diester diterpenoid alkaloids in the decoction presented mild changes,showing the decreasing sequence in mesaconitine,hypaconitine and aconitine.Moreover,the dissolution rule of diester diterpenoids alkaloids showed as the same as their contents,being increased firstly and then decreased obviously,and reached the maximum amount after 2 hours.Conclusion The quantitative method is feasible and precise,and can be used to illuminate the change rule of diester diterpenoids alkaloids in Poge Jiuxin decoction.
