摘要
目的:探讨人参皂甙肠道代谢物Compound K(CK)对人肝癌细胞MHCC97-H增殖及侵袭的影响,并初步探讨及可能的机制。方法噻唑蓝(MTT)比色法检测CK对细胞增殖活性的影响;流式细胞仪检测CK对MHCC97-H细胞周期的影响;Transwell小室法检测细胞侵袭能力;逆转录-聚合酶链反应及Western blot法分别检测p21、MMP-2及MMP-9 mRNA及蛋白的表达水平。结果与对照组相比,CK处理组的细胞增殖活性明显下降,细胞周期分析显示,随着CK浓度的增加,G0/G1期细胞所占百分率逐渐增加;CK作用肝癌细胞48 h后,侵袭穿膜细胞数分别为(22.24±7.3)、(18.24±4.6)个,与对照组比较差异有统计学意义(P〈0.01);RT-PCR及western blot结果显示CK能够促进p21的表达,下调MMP-2及MMP-9的表达。结论CK能够显著抑制人肝癌细胞的增殖和侵袭,机制可能与阻滞细胞周期以及影响相关基因表达有关。
ObjectiveTo investigate the effects of CK on proliferation and invasion of human liver cancer MHCC97-H cells, and appraise the possible mechanism.MethodsWith the treatment of CK in vitro, cell viability was assayed using MTT methods; Cell invasion of MHCC97-H cells were assessed by transwell technique. and the p21 and MMP-2/9 were detected by real-time RT-PCR and western-blot respectively.ResultsCompared with the control,CK decreased the proliferation of MHCC97-H cells, The colony formation ability of MHCC97-H cells was decreased and percentage of cells in G0/G1 phase was increased in a dose-dependent manner, as compared to control. The cell invasion abilities were decreased gradually. The real-time RT-PCR showed that p21 mRNA expression was increased and the MMP-2/9 mRNA was decreased.ConclusionCK inhibits the proliferation and invasion of MHCC97-H cells mainly by blocking the cell cycle at G0/G1 and down-regulating the expression of related gene factors.
