化瘀通络灸法对皮质下血管性痴呆大鼠的学习记忆能力及额叶皮质下组织病理变化的影响 被引量:6

摘要 目的:观察化瘀通络灸法对皮质下血管性痴呆(SVD)模型大鼠学习记忆能力和额叶皮质下形态学的影响。方法:4-VO法复制SVD大鼠模型。随机分为正常组、假模型组、模型组、药物组、艾灸组,每组10只。各组大鼠经治疗后跳台试验检测学习记忆能力,取材,光镜下观察各组大鼠大脑额叶皮质下组织形态学变化。结果:1大鼠跳台实验的潜伏期、错误次数,艾灸组及药物组与模型组比较差异具有统计学意义(P<0.01,P<0.05),艾灸组行为学成绩优于药物组(P<0.01,P<0.05)。2正常组与假模型组可见神经元排列整齐、密集,细胞核清晰,胞质丰富均匀;模型组神经元形态不规则,排序紊乱,细胞核固缩,核周及胞质内出现大小不同的空泡,胞质疏松,出现大量星形胶质细胞聚集增生;艾灸组细胞核染色较均匀,胞质形态良好,细胞排列较模型组整齐,形态接近正常组并可见少量星形胶质细胞和修复或新生少突胶质细胞;药物组可见坏死神经元,细胞排序不规则但较模型组整齐,细胞核染色略有不均,视野内可见少量星形胶质细胞增生。结论:化瘀通络灸法可提高SVD大鼠的学习记忆成绩,对SVD大鼠额叶皮质下损伤的神经元有修复作用。 Objective :To observe the effects of Huayutongluo Moxibustion on the function of learning and memory and morphology of frontal subcortical in subcortical vascular dementia(SVD)rats. Method:SVD rat models were established according to 4VO method. Randomly divided into the normal group,the model group,the sham model group,the medicine group and moxibustion group,10 rats in each group. The learning and memory ability of the rats in every group was assessed with jumping experiment after treatment,removed the material,observed the morphological changes of frontal subcortical of rats in each group under the light microscope. Result:① Statistica I difference existed in the latent period,the error times of jumping experiment in the moxibustion group and the medicine group when compared with the model group(P〈0.01,P〈0.05),and the moxibustion group was better than the medicine group(P〈0.01,P〈0.05). ② The normal group and sham model group showed neurons arranged in neat rows,the clear nucleus,rich and homogeneous cytoplasm. The model group showed neurons with irregular shape and sort disorder,karyopyknosis,different size of vacuoles appeared in the cytoplasm and nuclear around,the cytoplasm osteoporosis,and the emergence of a large number of astrocytes hyperplasia. The moxibustion group nuclear staining was uniform, cytoplasm in good shape. Compared with model group, cells arranged in neat,almost near the normal group.It is also showed a small amount of astrocytes and oligodendrocytes repair or newborn. The medicine group showed necrosis of neuron. The celling sorting of medicine group is irregular, but neater than model group. Nuclear staining was slightly uneven. It was showed a few astrocytes in the view..Conclusion:Huayutongluo Moxibustion can improve the ability of learning and memory in SVD rats observably,and repair the frontal subcortical injured neurons of SVD rats.
