NS398在大鼠胰十二指肠移植缺血再灌注损伤中对P选择素的影响

Influence of NS398 on P-selectin in rats with ischemia-reperfusion injury following pancreaticoduodenal transplantation

摘要 目的探讨NS398(一种特异性环氧合酶-2抑制剂)能否抑制大鼠胰十二指肠移植缺血再灌注损伤中P选择素(P-selectin,Ps)的表达。方法 60只SD大鼠随机分为3组:假手术组(12只)、移植组(12对)、移植+NS398组(12对)。三组分别于术后1、6、12h测定血清淀粉酶含量,胰腺组织学改变、胰腺组织中Ps的表达情况。结果各时间点:假手术组胰腺组织损伤不明显,血淀粉酶不升高,Ps不表达;移植组胰腺组织损伤加重,血淀粉酶升高,Ps明显表达;移植+NS398组胰腺组织轻度损伤,血淀粉酶轻度升高,Ps表达但明显低于移植组。结论 NS398能抑制胰十二指肠移植缺血再灌注损伤大鼠Ps的表达,对大鼠胰十二指肠移植缺血再灌注损伤有保护作用。 Objective To investigate whether NS398 can inhibit P-selectin in rats with ischemia-reperfusion injury following pancreaticoduodenal transplantation. Methods Sixty Sprague-Dawley rats were randomized into three groups. Sham operation group (ten rats), transplanted group (twenty-four rats)and transplanted with NS398 group (twenty-four rats). Blood samples in the three groups were harvested at 1,6 and 12 hours after the operation for serum amylase assay. Pancreas samples were harvested at the same time for morphological study by light microscopy. The expression of Ps protein of pancreas were analyzed immunohistochemical method, Results The level of serum amylase and the pancreas tissue had no marked change in each time spot of Sham operation group. As reperfusion was prolonged, the damage of pancreas tissue was aggravated and the level of serum amylase was elevated gradually in transplanted group, while these changes were mild in transplanted with NS398 group. There were no expressions of Ps in each time spot of Sham operation group. The expression of Ps in each time spot of transplanted and transplanted with NS398 group can be observed, but that in transplanted with NS398 group was conspicuous lower than that in transplanted group. Conclusion Under the ischemia-reperfusion injury following pancreaticoduodenal transplantation in rats, NS398 can inhibit the expression of PS and has protective effects.
