草菇丝氨酸蛋白酶基因在贮藏期表达变化的研究 被引量:1

摘要 采用不覆膜(control)与覆盖厚度为12μm、打6孔聚氯乙烯膜(PVC12-6)的方式分别对草菇进行保鲜贮藏,观察比较其蛋白酶活性,并通过实时荧光定量PCR技术研究草菇在贮藏期丝氨酸蛋白酶(SER)基因表达变化规律。结果表明:(1)control的草菇蛋白酶活性在储藏前期快速增加,在48h酶活性为3.81U/g·min,72h有所下降,但96h达到高峰,为5.80U/g·min;而PVC12-6的草菇蛋白酶活性在贮藏前期缓慢增加,在96h达到高峰,为2.76U/g·min,随后快速下降;在贮藏期间,control的草菇蛋白酶活性一直比PVC12-6高。(2)con-trol的丝氨酸蛋白酶基因表达量在48h达到最大,随后表达量下降较慢,且48h之后的表达量远高于24h的表达量;而PVC12-6的丝氨酸蛋白酶基因表达量在48h达到最高点后迅速下降,48h后的表达量远远低于24h的表达量。 The activity of protease of Volvariellavolvaceawhich were packaged by perforated packaging film were in?vestigated.The expression of the serine protease gene of the samples in different storage time were investigated by re?al-time quantitative PCR.The results showed that the fruit bodies with non-packaging retainedhighest activity of pro?tease which was 5.80U/g · min at 96h.Another sample reached thehighest activity of protease at 96h.The relative ex?pression of SER gene of all samples exhibited a rapid increase at 48h,then they were continuous decrease.But the rate of decreased expression in control was lower than another sample.This result was coincidence relation with the activity of protease.
