环氧合酶-2及细胞周期蛋白D1在腮腺肿瘤组织中的表达及相关性分析 被引量:4

摘要 目的探讨环氧合酶-2(COX-2)、细胞周期蛋白D1(cyclinD1)在腮腺肿瘤组织中的表达及相关性。方法对河北联合大学附属医院曹妃甸区医院2009年3月-2014年9月收治的25例腮腺多形性腺瘤、42例癌在多形性腺瘤中及20例腺瘤旁组织(对照)病理标本进行免疫组织化学SP法染色,观察COX-2、cyclinD1的阳性表达情况及两者的相关性。结果癌在多形性腺瘤中、腮腺多形性腺瘤、腺瘤旁组织中COX-2的阳性率分别为83.3%、52.0%、5.0%,腮腺多形性腺瘤、腺瘤旁组织的COX-2阳性率与癌在多形性腺瘤中比较,差异均有统计学意义(均P〈0.05),腮腺多形性腺瘤与腺瘤旁组织比较,差异有统计学意义(P〈0.05)。癌在多形性腺瘤中、腮腺多形性腺瘤、腺瘤旁组织中cyclinD1的阳性率分别为81.0%、44.0%、10.0%,腮腺多形性腺瘤、腺瘤旁组织的cyclinD1阳性率与癌在多形性腺瘤中比较,差异均有统计学意义(均P〈0.05),腮腺多形性腺瘤与腺瘤旁组织比较,差异有统计学意义(P〈0.05)。在癌在多形性腺瘤中,COX-2及cyclinD1的表达与年龄、性别、肿瘤直径无关(P〉0.05);cyclinD1的表达与TNM分期及淋巴结是否转移有关(P〈0.05),COX-2的表达仅与TNM分期有关(P〈0.05)。在腮腺肿瘤中COX-2与cyclinD1的表达呈正相关(r=0.812,P〈0.01)。结论 COX-2、cyclinD1随着肿瘤的发生及恶化,其表达均逐渐增高,且两者的表达呈正相关性,提示COX-2、cyclinD1与腮腺肿瘤的发生、发展密切相关。COX-2、cyclinD1的联合检测有助于对肿瘤良恶性的鉴别,可对治疗方案选择提供参考。 Methods The expression of COX-2, cyclinD1 were detected by SP immunohistochemical staining in specimens of 25 cases of parotid pleomorphic adenoma, 42 cases of carcinoma in pleomorphic adenoma and 20 cases of tissues beside the adenoma(control). The positive expressions and correlation of COX-2, cyclinD1 were observed. Results The positive expression rate of COX-2 in carcinoma in pleomorphic adenoma, pleomorphic adenoma of parotid gland, tissues beside the adenoma were 83.3%, 52.0%, 5.0% respectively. The positive expression rate of COX-2 in carcinoma in pleomorphic adenoma was significantly higher than that in pleomorphic adenoma of parotid gland, tissues beside the adenoma(P〈0.05); the positive expression rate of COX-2 in pleomorphic adenoma of parotid gland was significantly higher than that in tissues beside the adenoma(P〈0.05). The positive expression rate of cyclinD1 in carcinoma in pleomorphic adenoma, pleomorphic adenoma of parotid gland, tissues beside the adenoma were 81.0%, 44.0%, 10.0%,respectively. The positive expression rate of cyclinD1 in carcinoma in pleomorphic adenoma was significantly higher than those in pleomorphic adenoma of parotid gland, tissues beside the adenoma(P〈0.05); the positive expression rate of cyclinD1 in pleomorphic adenoma of parotid gland was significantly higher than that in tissues beside the adenoma(P〈0.05). In carcinoma in pleomorphic adenoma, the expression of COX-2 was only associated with TNM staging(P〈0.05), the expression of cyclinD1 was associated with the lymph node metastasis, TNM staging(P〈0.05); both of the expressions of COX-2 and cyclinD1were not associated with the patient's gender and age, the size of tumor(P〉0.05). There was a positive correlation between the expression of COX-2 and the expression of cyclinD1 in parotid tumor(r = 0.812, P 0.01). Conclusion The expressions of COX-2, cyclinD1 are gradually increased with occurrence and progression of parotid gland tumor. There is a positive correlation between the expression of COX-2 and cyclinD1. The combined detection of COX-2 and cyclinD1 can help to differentiate benign and malignant tumors and help for antitumor therapy selection.
