乳酸盐脱氢酶及糖酵解活性与多发性骨髓瘤化疗耐受性的相关性研究

Study on the Correlation Between Lactate Dehydrogenase and Glycolytic Activity and Multiple Myeloma Chemotherapy Tolerance

摘要 目的探讨能量代谢异常与多发性骨髓瘤化疗耐受性的相关性。方法选取2010-05-2014-05间我院收治的48例多发性骨髓瘤患者为研究对象,按治疗效果分为CR组、PR组及NR组,选取同期于我院行健康体检的31例健康者为对照组,比较各组血清乳酸盐脱氢酶(LDH)水平。体外培养多发性骨髓瘤细胞株RPMI8226及相对应的马法兰耐药细胞株RPMI8226/LP,采用葡萄糖消耗实验检测各细胞株糖酵解活性。结果多发性骨髓瘤患者血清LDH均显著高于对照组,且血清LDH水平在CR组、PR组及NR组中呈逐渐升高趋势,各组比较差异具有统计学意义(P〈0.05)。多发性骨髓瘤细胞株RPMI8226的相对葡萄糖耗氧量为(71.5%±2.2%),其相对应的马法兰耐药细胞株RPMI8226/LP的相对葡萄糖耗氧量为(99.7%±0.7%),两组比较差异具有统计学意义(P〈0.05)。结论能量代谢水平与多发性骨髓瘤化疗耐受性存在相关性,调节机体能量代谢水平可能是改善多发性骨髓瘤化疗耐受性的新思路。 Objective To investigate the correlation of abnormal energy metabolism with multiple myeloma chemotherapy tolerance.Methods Selecting 48 cases with multiple myeloma from May,2010 to May,2014 in the hospital as the research object,dividing them into CR,PR and NR groups according to the treatment effect,selecting 31 healthy subjects as the control group. The comparison of serum lactate dehydrogenase of different groups was done. In vitro cultivation of multiple myeloma cell line RPMI8226 flange drug- resistant cell line and related horse RPMI8226 / LP,glucose consumption experimental detection was done by using the cell line activity of glycolysis. Results Multiple myeloma patients were significantly higher than that of control group,serum LDH and serum LDH level in CR,PR group and NR group had a rising trend,and differences of each group were statistically significant( P〈0. 05). The relative oxygen consumption of glucose to multiple myeloma cell line RPMI8226 was( 71. 5% ± 2. 2%),and relative oxygen consumption of glucose to its corresponding horse flange drug- resistant cell line RPMI8226 / LP was( 99. 7% ± 0. 7%),and the difference of the two groups was statistically significant( P〈0. 05). Conclusion There is tolerance correlation of the level of energy metabolism and multiple myeloma chemotherapy,and regulating the body's energy metabolism may be new ideas to improve multiple myeloma chemotherapy tolerance.
