摘要
目的建立高抵抗素血症的高血压大鼠模型,观察提高血清中抵抗素水平对自发性高血压大鼠血压水平的影响。方法将36只10周龄高血压大鼠(雄性),随机平均分为病毒组及对照组。其中病毒组经尾静脉注射载有抵抗素基因的重组腺病毒1x10PFu/mouse(用生理盐水稀释至0.3 ml)后随机平均分成第一周组、第四周组、第八周组;而对照组则连续经尾静脉注射等体积生理盐水后也随机平均分为第一周组、第四周组、第八周组。采用无创尾动脉仪测量各组大鼠的血压,同时采用竞争性酶联免疫吸附试验法检测各组大鼠血清抵抗素水平。结果病毒组与对照组比较,病毒组的收缩压及舒张压水平高于对照组,以第一周及第四周有统计学差异,其中,以第四周血压升高为更明显(P<0.05);病毒组的血清抵抗素水平较对照组升高,以第一周及第四周有统计学差异,其中,第四周升高幅度明显(P<0.05);抵抗素与收缩压(r=0.639,P=0.000),舒张压(r=0.613,P=0.000)均呈正相关。结论:血清抵抗素水平可促进高血压大鼠血压的升高,提示抵抗素可能在高血压的发生、发展过程中起一定的作用。
Objective: To establish the animal model of hyperresistinemia and to observe the effects of resistin on blood pressure. Methods: 36 male spontaneous hypertension rats who were 10 week old were randomly divided equally into virus group and control group. Virus group wered injected recomninant adenovirus of resistin gene through cauda vein( 1x10 PFu /mouse). Then the virus group were randomly divided into the first week group,the fourth week group and the eighth week group. the control group were injected an equalent volume of saline through cauda vein,Then the control group were randomly divided into the first week group,the fourth week group and the eighth week group. we measured blood pressure and serum resistin. Results: Results systolic blood pressure and diastolic blood pressure in the virus groups were increased; the first week group and the fourth week group systolic blood pressure and diastolic blood pressure were higher than the control group. The fourth week group blood pressure were increased obviously( P〈 0. 05). The serum resistin levels were higher in the virus groups; the difference between the first week group and the fourth week group were statistically significant( P〈 0. 05). The serum resistin of the fourth week group was higher than the first week group. Pearson correlation analysis revealed their systolic blood pressure( r = 0. 639,P = 0. 000) and diastolic blood pressure( r = 0. 613,P = 0. 000) were directly correlated with the serum resistin. Conclusion: serum resistin increased blood pressure in rats and possibly plays a pivotal role in pathogenesis and hypertensive development.
