摘要
本试验旨在研究不同剂量的苦马豆素(SW)、黄花碱(Ts)及其2类生物碱的混合物胃内灌服小白鼠,检测血液中T淋巴细胞的增殖性反应。将240只小白鼠随机分组,分别灌服不同浓度的SW、Ts和两类生物碱的混合物,并于试验的第7、14、21、28、35d每组随机抽取4只小白鼠,眼球采抗凝血,分离淋巴细胞,用Con A刺激,MTT法检测T淋巴细胞增值性反应情况。结果显示,低剂量的SW能够提高T淋巴细胞增殖,而中等剂量和高剂量短时间内对T淋巴细胞的增殖也有促进作用,长时间会严重的抑制T淋巴细胞的增殖;低剂量的Ts对T淋巴细胞的增殖几乎无影响,高剂量也会抑制T淋巴细胞的增殖;混合物出现了与SW类似的结果。结果表明,不同浓度的SW、Ts及其混合物均能不同程度的影响T淋巴细胞的增值性反应。
This study is designed to discuss the mice were drenched different doses of swainsonine( SW),Thermopsine( Ts) and the mixture of two alkaloids,then detection of proliferative response of T lymphocytes in the blood. 240 mice were randomly divided into three groups and were drenched different concentrations of SW,Ts and the mixture of two alkaloids respectively. Each group was randomly selected four mice on the 7th,14 th,21st,28 th,35th day and the mice were taken eye anticoagulant to isolate lymphocytes.Lymphocytes were stimulated by Con A and the T lymphocyte transformation was detected by MTT assay. The result showed that low-dose SW could improve the proliferation of T lymphocytes and middle- dose and high- dose SW could also improve the proliferation of T lymphocytes in short time,but they could seriously reduce the proliferation of T lymphocytes in long time. Low- dose Ts almost had no effect on the proliferation of T lymphocytes,but high- dose Ta also could seriously reduce the proliferation of T lymphocytes. The mixture of two alkaloids appeared similar results with SW. The result showed that the different concentrations of SW,Ts and the mixture of two alkaloids can cause somewhat impacts on T lymphocyte proliferative response.
