摘要
以大豆绥农28为材料,用发根农杆菌(Agrobacterium rhizogenes)K599浸染转化后诱导产生发根,通过β-葡聚糖苷酶(GUS)染色进行效率检测,确定毛状根转化的最佳条件。结果表明绥农28大豆品种发根诱导高效转化的条件如下:在1/2MSB5+20g/L蔗糖+琼脂7g/L,p H5.8萌发培养基上萌发5~6d的大豆子叶节为外植体,当菌液侵染时间为20min,共培养基p H5.4的条件时,发根转化率为58.57%。
Hair roots of Soybean Suinong 28 was induced by Agrobacterium rhizogenes K599,then the efficiency of GUS staining of hairy roots were detected,which determined the optimum conditions for transformation of hairy root. The results showed that the best conditions for transformation on soybean Suinong 28 were: germination medium in the 1 /2MSB5 + sucrose 20 g / L + agarose 7g / L,p H5. 8; Soybean Cotyledonary node as explants on the 5- 6 days; infected by the bacteria for 20 minutes; in the co- culture medium p H5.4 for 5 days; hair roots was induced on medium of 1 /2MSB5 + sucrose 30 g / L + agar 7g / L,p H5. 8. Under the conditions,rate of transformation of hair roots was 58. 57%.
