凋亡调控因子c-IAP1,c-IAP2和Caspase-3在大肠肿瘤中的表达及意义

摘要 目的:探讨凋亡调控因子c-IAP1,c-IAP2和Caspase-3在大肠肿瘤中的表达,了解其在大肠肿瘤的发生发展中的作用。方法:收集我院2002年至2004年手术后经病理证实并有完整随访资料的大肠腺癌标本45例,同时收集外伤后手术切除的正常大肠组织为正常对照,采用免疫组化SP法和半定量积分法分别检测c-IAP1,c-IAP2和Caspase-3在大肠腺癌、大肠腺瘤及正常大肠黏膜中的表达。结果:c-IAP1,c-IAP2和Caspase-3的阳性表达率在正常大肠黏膜组与大肠腺瘤组、正常大肠黏膜组与大肠腺癌组、大肠腺瘤组与大肠腺癌组之间均有显著性差异(P<0.05)。c-IAP1与Caspase-3在大肠腺癌中的表达呈负相关(r=-0.321,P<0.05)。c-IAP2和Caspase-3在大肠腺癌中表达无相关性,(P>0.05,r=-0.164)。c-IAP1和c-IAP2在大肠腺癌中的表达无相关性。(P>0.05,r=0.066)。结论:凋亡受阻在大肠肿瘤形成过程中起着一定的作用;c-IAP1,c-IAP2可能参与了大肠腺瘤和腺癌的发生和发展,并在一定程度上反映大肠腺癌的恶性程度;凋亡抑制蛋白c-IAP1,c-IAP2的表达与患者的性别、年龄、5年生存率、肿瘤部位、组织学类型、有无淋巴结转移及Dukes分期等因素无关。 Objective To investigate the expressions of c-IAP1, c-IAP2 and Caspase-3 and their clinical pathological characteristics in colorectal carcinoma (CRC). Methods 45 cases of CRC pathological verified from January 2002 to January 2004 with surgical resection were selected and the expressions of c-IAP1, c-IAP2 and Caspase-3 were detected by immuno-histochemical SP and Semi quantitative weighted integral methods. Results Pairwise comparison showed significantly different expression rates between normal and adenoma group, normal and CRC group, adenoma and CRC group(P〈0.05). Pairwise comparison showed c-IAP1 was associated with Caspase-3 in CRC(r = -0.321, P 〈 0.05),c-IAP2 was not associated with Caspase-3 in CRC(P 〉 0.05, r = -0.164), c-IAP1 was not associated with c-IAP2 in CRC (P 〉 0.05, r = 0.066). Conclucions Apoptosis inhibition was associated with the occurrence of CRC. C-IPA1, c-IAP2 may took part in the differentiation of CRC. C-IPA1, c-IAP2 reflected the degree of malignancy of CRC. C-IPA1, and c-IAP2 was not associated with sex, age, survival period, tumor side, histological type, lymph node metastasis and Dukes stages.
