摘要
目的:利用基因芯片技术,研究miRNA在喉鳞癌和癌旁正常组织中的表达水平差异,寻找可能的分子生物学指标。方法:8例经手术的喉鳞癌及其周边(>1cm)的正常组织,以Trizol法提取总RNA,经Cy3荧光标记后与含有人723条miRNA探针的芯片进行杂交,筛选在喉鳞癌及周边正常组织中差异表达的miRNA。结果:共有132个miRNA在8例喉鳞癌中均有表达。喉鳞癌和癌旁正常组织中差异表达的miRNA有53个,其中22个出现上调,31个出现下调,差异表达5倍以上的miRNA共有8个,分别为miR-196b、miR-196a、miR-375、miR-139-5p、miR-7、miR-338-3p、miR-183和miR-370。其中,miR-196b、miR-196a和miR-375差异表达8倍以上。结论:通过基因芯片的方法,发现了喉鳞癌中53个异常表达的miRNA,其中8个miRNA有明显差异的表达,在这些差异表达的miRNA中寻找可能存在的靶基因,为进一步探索喉鳞癌发病机制奠定基础。
Objective To study differential expression of microRNAs between carcinoma and adjacent normal tissue in squamous cell larynx cancer, and to find the potential molecular biomarkers. Methods Total RNA of tumor and normal tissues was extracted from 8 patients with squamous cell laryngeal carcinoma by Trizol method, and was labeled with cyanine3-pCp and hybridized with human 723 miRNA mieroarray. Differential expression of miRNA between tumor and normal tissues was determined. Results 132 miRNAs were determined in all 8 samples, and 53 of them were found to have differential expression. 22 of the 53 showed up-regulated, and others down-regulated. 8 miRNAs were found to have more than 5-fold differential expression, which were miR-196b, miR-196a, miR-375, miR- 139-5p, miR-7, miRo338-3p, miR-183 and miR-370. Among them, miR-196b, miR-196a and miR-375 showed more than 8-fold expression in laryngeal carcinoma. Conclusions 53 miRNAs have differential expression by microarray method, and 8 of them show significant differential expression. The study of the differential miRNAs between laryngeal carcinomas and normal tissues and their putative target genes may play a fundamental role for the further studies in squamous cell laryngeal carcinoma.
