电针、埋线调整吗啡成瘾小鼠SCN内Per基因表达的研究 被引量:4

摘要 目的:探讨电针、埋线对吗啡成瘾小鼠大脑视交叉上核(suprachiasmatic nucleus,SCN)内生物钟基因Per1、Per2表达的影响。方法:筛选雄性C57BL/6J近交系小鼠42只,随机分为空白组(n=14)、模型组(n=14)、电针组(n=7)、埋线组(n=7)。动物模型根据"7 d递增成瘾法"皮下注射盐酸吗啡注射液。第7天取小鼠SCN,采用实时荧光定量PCR法检测Per1、Per2基因的表达水平。结果:与空白组小鼠比较,模型组小鼠SCN内Per1、Per2基因表达均下调,差异具有统计学意义(P<0.05);与模型组小鼠比较,电针组Per2基因表达明显上调(P<0.05),埋线组Per1基因表达明显上调(P<0.05);电针组与埋线组比较,差异无统计学意义(P>0.05)。结论:埋线和电针可以分别调节Per1、Per2基因的表达,从而改善吗啡成瘾小鼠紊乱的节律。 Objective:To discuss the effects of electro-acupuncture and catgut embedding in regulating expressions of Per1 and Per2 in suprachiasmatic nucleus(SCN)of morphine-addiction mice. Methods:42 male C57BL/6J mice were randomly divided into blank group(n=14),model group(n=14),electro-acupuncture group(n=7)and catgut embedding group(n=7). Models were induced by injecting morphine hydrochloride injection. Samples of SCN were removed and Per1 and Per2 genes expression were measured by RT-PCR method. Results:Compared with blank group,gene expressions of Per2 and Per1 were up-regulated in model group(P〈0.05). Compared with model group,Per1 expression of catgut embedding group and Per2 expression of electro-acupuncture group were up-regulated(P〈0.05). Conclusion:Therapies of catgut embedding and electro-acupuncture could improve indiscriminate rhythm of morphine dependent mice through regulating expressions of Per1 and Per2.
