摘要
目的:探讨载脂蛋白A5(Apo lipoprotein A5,APOA 5)基因多态性与血脂和颈动脉粥样硬化的关系。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法检测154例颈动脉粥样硬化患者(颈动脉硬化组)和150例健康对照者(对照组)载脂蛋白A5的-1131T>C基因型的分布。通过颈动脉彩色多普勒超声检查测量颈总动脉(CCA)和颈内动脉(ICA)内膜中层厚度(IMT)评估颈动脉粥样硬化程度。结果:颈动脉硬化组中基因型为TT、TC、CC三种,-1131CC基因型和-1131TT+TC基因型人群的CCA厚度和ICA厚度间的差异无统计学意义(P>0.05);颈动脉硬化组ApoA 5基因-1131T/C位点C等位基因携带者(TC基因型和CC基因型)的血清TG水平和TG/HDL-C比值均高于TT基因型携带者(P<0.05)。对照组TC基因型携带者的血清TG水平和TG/HDL-C比值较TT基因型携带者升高(P<0.05)。结论:ApoA 5基因-1131T/C多态性与颈动脉粥样硬化的发生可能具有相关性,其中CC基因型患颈动脉粥样硬化的风险最大。ApoA 5基因-1131T/C多态性对血清TG的水平有重要的影响。
Objective To investigate the role ofAPOA5-1131 gene T/C polymorphism in blood lipid and carotid atherosclerosis. Methods The distribution of Apo lipoprotein A5 genotype 1131 T/C in 154 patients with carotid atherosclerosis (carotid atherosclerosis group) and 150 healthy controls (control group) was analyzed by polymerase chain reaction restriction fragment length polymorphism assay. The atherosclerosis degree of common carotid artery (CCA) and internal carotid artery (ICA) intima-media thickness (IMT) was evaluated by carotid ultrasound. Results The genotype of carotid atherosclerosis was TT, TC and CC, no significant differences in -1131CC and -1131TT+TC genotypes of the CCA population between the thickness and ICA thickness (P 〉 0.05) was found; The level of serum TG and TG/HDL-C ratio were higher than that of TT genotype carriers (P 〈 0.05). Control group TC genotype carriers of serum TG level and TG/HDL-C ratio than TT genotype carriers increased (P 〈 0.05). Conclusion The result shows that there is correlation between the APOA5-1131 gene T/C polymorphism and carotid atherosclerosis, and that the CC genotype has the greatest risk to develop carotid atherosclerosis. Carotid atherosclerosis group APOA5-1131 gene T/C locus in different genotype subgroups of carotid artery IMT shows no statistical significance (P 〉 0.05).
