阿托伐他汀降脂作用与APOA5基因多态性相关性分析

摘要 目的:研究分析Apo A5-1131bp T>C基因多态性与阿托伐他汀降脂作用的相关性。方法:选取2012年2月-2013年6月神经内科和心血管内科住院的血脂异常患者324例,采用多聚酶链反应-限制性片段长度多态性法进行基因型检测,并随机选取其中87名患者,口服阿托伐他汀10 mg/d,分析比较Apo A5-1131bp T>C各基因型的服药前后的血脂水平,同时收集所研究患者各项实验室指标、流行病学问卷等,并对所有研究数据进行统计分析。结果:服药前甘油三酯(TG)水平TC型组与CC型组明显高于TT型组,而以CC型组的TG水平最高,组间差异具有统计学意义(P<0.05);服药12周后发现C携带者的TG降低水平明显高于非C携带者,组间差异具有统计学意义(P<0.05),其余的血脂指标则没有此项发现。结论:Apo A5-1131bp T>C基因启动区基因多态性与血浆血脂水平存在一定关联,该位点T/C转换与高血脂有关。 Objective To investigate the relationship of the polymorphism of Apo A5-1131 T C antihyperlipidemia effi cacy response to Atorvastatin treatment in patients with essential hypertension. Methods A total of 324 essential hyperlipoidemia patients were selected from China and treated with Atorvastatin for a period of 12 weeks.The Apo A polymorphisms were genotyped using polymerase chain reaction-restriction fragment length polymorphism method. In 324 patients who had been measured blood lipid at the beginning and ending. Epidemic information and physical related data were also collected. The data were analyzed by the methods of ANOVA and multiple linear regressions. Results Apo A5-1131 T 〉C genotypes of people living with a signifi cant difference in the level of TG,TC and CC-type group was signifi cantly higher than the TT-based group,and CC-type had the highest level of TG(P〈0.05);medication after 12 weeks,TG showed the same trends and changes in medication as before. Medication before and after comparison found only in patients with genotype CC TC and LDL-C level dropped signifi cantly higher than that of TC Group and the TT group(P〈0.05),the rest of the lipid indicators were not found the same results. Conclusion APOA5-1131 bp T〉 C gene promoter region polymorphism has relationship with plasma lipid levels and there is a certain relevance, which is related to high blood lipid conversion;the effect of atorvastatin shows descending order for TT group, TC group and CC group.
