摘要
目的:研究左归丸胚胎期干预能否对妊娠糖尿病(GDM)大鼠子代高糖高脂饮食诱发的糖耐量减低(IGT)起到预防作用。方法:育龄雌性SD大鼠受孕后随机分为正常对照组、妊娠糖尿组、胰岛素组、左归丸组、联合干预组。受孕当日妊娠糖尿病组和3个干预组按45 mg/kg腹腔内注射链尿佐菌素(STZ),复制妊娠糖尿病模型。并于孕4 d^19 d给药干预(左归丸10 g/kg灌胃,胰岛素20 IU/kg皮下注射)。孕鼠产子后,每窝取1只雄鼠,组成新组,组别不变。母乳喂养3 w,普通生长饲料喂养2 w,除正常对照组外,其余各组子鼠均于6 w龄开始高糖高脂饮食。10 w后葡萄糖干化学法测各组子鼠空腹血糖(FPG)、糖负荷后2 h血糖(P2h G),全自动生化仪测血脂,酶联免疫法测脂联素浓度,并进行HE染色观察肝脏的病理形态。结果:15 w龄末,模型组子鼠发生IGT;与正常对照组比较,妊娠糖尿病组TC、TG显著升高,HDL-C、APN显著降低,药物干预组可以改善相关指标,联合用药组优于单用药物组。结论:糖尿病大鼠妊娠期予补肾阴方左归丸干预能够有预防高糖高脂饮食引发子代成年大鼠发生IGT。
Objective:To study the preventive effect of Zuogui Pill on rats with GDM offspring induced by itigh-fat diet. Methods:Pregnant reproduction SD rats were randomly divided into normal pregnancy group, GDM control group, diabetes insulin treated group,Zuogui Pill treated group,Zuogui Pill combined with insulin treated group.Sreptozotocin (STZ)were injected in rats abdomen so as to induce GDM model in GDM control group and the three treated groups. GDM control group and the three treated groups were prevented by drug respectively. After offsrings were born,a male mouse were tak- en to form a new group. The mice were given breastfeeding 3 weeks and general growth of diet 2 weeks. Except normal control group,other groups were given the high fat diet after 6 weeks. FPG and P2hG were measured by dry chemical measurement of glucose, blood lipids were measured by ACA, adiponectin concentration were measured by ELISA and liv- er pathomorphism were stained by HE and observed. Results:At the end of 15 weeks,IGT model were occurred in the model group offspring. Compared with the normal group, model group TC, TG significantly increased, HDL-C, APN signifi- cantly reduce. Conclusions : Zuogui Pill prevention in the period of diabete rats pregancy has the prevention offspring adult rats effect of occurrence of IGT induced by high-sugar and high-fat diet in adult rats
