摘要
为探究油菜蜂花粉黄酮抗糖尿病及其并发症的作用,利用α-葡萄糖苷酶抑制实验、四唑氮蓝(nitroblue tetrazolium,NBT)还原实验、二羰基化合物测定和非酶糖基化高级终产物荧光强度测定等方法,研究了油菜蜂花粉黄酮对α-葡萄糖苷酶及非酶糖基化反应的抑制作用。结果表明:油菜蜂花粉黄酮对α-糖苷酶的半抑制质量浓度为0.072 16g/L,远小于阳性对照阿卡波糖的,抑制类型为混合型;对非酶糖基化反应的3个阶段均具有抑制作用,且抑制率都随黄酮浓度的增大而增大,其中对第3阶段的抑制作用最强,黄酮质量浓度为0.227 0g/L时,3个阶段的抑制率分别为25.92%、39.24%、59.45%。实验证实,油菜蜂花粉黄酮可在降低α-葡萄糖苷酶活性的同时,抑制非酶糖基化反应的发生。
In order to explore the resistance to diabetes and its complications of flavonoids in Brassica napus L.bee pollen,the inhibitory effect onα-glucosidase and the advanced glycation end products(AGEs)formation were investigated through enzyme inhibition test,nitroblue tetrazolium(NBT)reduction assay,measurement of dicarbonyl compounds and the fluorescence of AGEs.The results showed that flavonoids in B.napus L.bee pollen had inhibitory effect onα-glucosidase and all the three periods of AGEs formation.The IC50 of flavonoids onα-glucosidase was 0.072 16g/L,which was much less than the positive of acarbose.The kinetic analysis showed that the flavonoids behaved as a mixed-type inhibitory mechanism.For the three periods of AGEs formation,inhibition rate had positive correlation with concentration of total flavonoids.Especially,it had the strongest inhibitory effect on the third stage.When the total flavonoids concentration was 0.227 0g/L,the three stages inhibition rates on non-enzymatic glycosylation reaction were 25.92%,39.24% and 59.45%,respectively.Above results confirmed that the flavonoids in B.napus L.bee pollen had inhibitory effect onα-glucosidase and AGEs formation.
