X染色体连锁凋亡抑制基因小分子抑制剂Embelin对宫颈癌细胞株HeLa细胞凋亡的影响及分子机制 被引量:3

摘要 目的:研究X染色体连锁凋亡抑制基因(XIAP)小分子抑制剂Embelin在体外对宫颈癌细胞株He La生长的影响,并探讨其作用机制。方法:应用CCK-8检测不同浓度Embelin对宫颈癌细胞株He La增殖的影响;利用流式细胞仪检测Embelin对He La细胞凋亡的影响;Western blot方法检测Embelin作用后细胞XIAP、Caspase-3、Caspase-9表达的变化。结果:Embelin对宫颈癌细胞株He La增殖具有显著抑制作用,差异有统计学意义(P<0.05);经不同浓度Embelin处理后,He La凋亡率明显增高,与对照组比较差异有统计学意义(P<0.01);经Embelin作用24 h后,He La细胞Caspase-9,Caspase-3,PARP被激活,表达增加。结论:Embelin在体外可显著抑制He La细胞增殖,诱导细胞凋亡,其机制可能是通过拮抗XIAP的作用,激活Caspase依赖性的细胞凋亡途径。 Objective: To study the effect of X-linked inhibitor of apoptosis (XIAP) Embelin on growth of cervical cancer HeLa cells in vitro, explore the mechanism. Methods: CCK-8 was used to detect the effects of different concentrations of Embelin on proliferation of cervical cancer HeLa cells; flow cytometry was used to detect the effect of Embelin on apoptosis of cervical cancer HeLa cells ; Western blot was used to detect the changes of XIAP, Caspase-3, and Caspase-9 expressions in cervical cancer HeLa cells after treated with Embelin. Results: Embelin significantly inhibited the proliferation of cervical cancer HeLa cells ( P〈0.05 ) . The apoptotic rate of cervical cancer HeLa cells significantly increased after treated with different concentrations of Embelin, compared with control group, there was statistically significant difference (P〈0.01 ) . Treating cervical cancer HeLa cells with Embelin after 24 hours, Caspase-9, Caspase-3, and PARP were activated, and their expressions increased. Conclusion: Embelin can significantly inhibit proliferation and induce apoptosis of cervical cancer HeLa cells in vitro, which may take effect by antagonizing XIAP and activating Caspase-dependent apoptotic pathway.
