艾灸三阴交、关元对寒凝证类痛经大鼠子宫收缩强度和OTR的影响 被引量:16

Effect of Moxibustion at "'Guanyuan'(RN4)" and " 'Sanyinjiao'(SP6)" on the Tension of the Uterus and OTR Expression on Dysmenorrheal Rat Model with Cold Stagnation Syndrome

摘要 目的:观察艾灸三阴交、关元穴对寒凝证类痛经大鼠子宫收缩强度及子宫中缩宫素受体(OTR)的影响,旨在初步探讨艾灸不同穴位对寒凝证类痛经大鼠镇痛效应的差异及可能的分子响应机制。方法:将处于动情间期3月龄的SD雌性大鼠40只,随机分为盐水组、寒凝类痛经组、三阴交组和关元组,每组10只。除盐水组外,寒凝类痛经组、三阴交组和关元组采用全身冷冻法结合苯甲酸雌二醇注射法造模。盐水组和寒凝类痛经组不予艾灸,三阴交组在造模第6天、第8天注射苯甲酸雌二醇后及在第10天注射缩宫素后,在大鼠三阴交穴(双侧)予以艾柱施灸,每次灸3壮。关元组灸治方法与之相同。运用BL-420E+生物机能实验系统检测在体大鼠子宫收缩强度,并用RT-PCR法测定大鼠子宫中OTR的表达水平。结果:1.与盐水组比较,寒凝类痛经组子宫收缩强度明显增强(P<0.01);与寒凝类痛经组比较,两艾灸组均显著减少了子宫收缩波个数(P<0.01),且降低了子宫活动度(P<0.05)。2.与盐水组比较,寒凝类痛经组子宫OTR的峰值增高(P<0.01);与寒凝类痛经组比较,三阴交组和关元组子宫OTR的峰值、均值、泳道值均显著降低(P<0.01)。结论:(1)艾灸可明显减轻大鼠的类痛经反应,缓解子宫痉挛性收缩。(2)本实验结果显示两穴镇痛效应未见明显差异。 Objective: To investigate the differences in analgesic effect and the probably molecular response mechanism of the dysmenorrheal rat model with cold stagnation syndrome by moxibustion at "'Guanyuan'(RN4)" and"'Sanyinjiao'(SP6)", which by observing the writhing reaction and the OTR expression in uterus .Methods: The 40 anestrous 3-month-old female SD rats were divided into four groups according to the random number(n=10),including saline group, cold stagnation Of primary dysmen- orrheal model group, moxibustion "'Sanyinjiao'(SP6)" group and moxibustion "'Guanyuan'(RN4)" group. Other groups except saline group to preparation of cold stagnation of primary dysmenorrheal model through combine systemic freezing and injection estradiol benzoate. Did not implemented moxibustion to the saline group and the cold stagnation of primary dysmenorrheal model group. The moxibustion "'Sanyinjiao'(SP6)" group treated on the sixth day and the eighth day after injected estradiol benzoate, as well as on the tenth day after injected oxytocin. Both side "'Sanyinjiao'(SP6)" were implemented moxibustion, 3 cones at each point every time. The moxibustion "'Guanyuan'(RN4)" group was treated with the same method. Using BL-420E + biological and functional experimental system to detect the intensity of the contraction in the rat uterus,and using the method of RT-PCR to detect OTR expression. Results:l.Compared with the saline group, model group significantly increased the intensity of uterine contractions(P〈0.01); Compared with the model group,Two moxibustion groups both significantly reduced the number of uterine contraction waves (P〈0.01), and reduced uterine activity(P〈0.05). 2.Compared with the saline group, the cold stagnation of primary dysmenorrheal model group increased the peak of uterine OTR; Compared with the the cold stagnation of primary dysmenorrheal model group, Two moxibustion groups both obviously lowed the peak, mean and lane values of uterine OTR(P〈0,01). Conclusion:(1)Moxibustion can significantly reduce dysmenorrheal response in rats and relieve spastic contraction of the uterus. (2)The experiment results show that the two holes analgesic effect was no significant difference.
