口腔鳞癌中β-catenin和p53蛋白的表达及其临床意义 被引量:3

摘要 目的研究口腔鳞癌组织中β-catenin和p53蛋白的表达及其与临床病理学特征的关系,探讨β-catenin和p53蛋白在口腔鳞癌发生中的作用。方法采用免疫组织化学方法检测55例原发性口腔鳞癌组织以及15例正常口腔黏膜组织中β-catenin和p53蛋白的表达。结果 (1)β-catenin在口腔鳞癌组织中显示细胞膜表达减弱或消失,并出现胞质和(或)胞核内的异常蓄积。β-catenin在口腔鳞癌组织中的异常表达率为70.9%,明显高于正常对照组(P<0.05),而且在口腔鳞癌中-低分化型的异常表达明显高于高分化型,与肿瘤的组织分化程度有关(P<0.05),但与有无淋巴结转移差别无统计学意义(P>0.05)。(2)p53蛋白的阳性表达率为67.2%,显著高于正常口腔黏膜组织(P<0.05)。p53蛋白在口腔鳞癌临床Ⅲ、Ⅳ期的过度表达明显高于Ⅰ、Ⅱ期,差异有统计学意义(P<0.05)。在有无淋巴结转移组间p53蛋白的阳性表达差异无统计学意义(P>0.05)。(3)β-catenin异常表达与p53蛋白阳性表达之间呈正相关(r=0.373,P<0.05)。结论β-catenin和p53蛋白单独或可能协同作用参与口腔鳞癌的发生发展。 Objective To study the expression of β - catenin and p53protein in oral squamous cell carcino- ma (OSCC) and their clinical significance. Methods Immunohistochemical analysis was applied to detect the expressions of β- catenin and p53 protein in 55 cases of oral squamous cell carcinoma and 15 cases of normal oral mucous membrane. Results (1) Abnormal expression of β - catenin was described as reduced or absent mem- brane staining and altered cytoplasmic and/or nuclear accumulation and was detected in70.9 % of 55 cases in OS- CC. Abnormal β - catenin expression was significantly associated with moderately or poorly differentiated carci- noma(P 〈 0.05), however, there was no a statistically significant association with lymph node metastasis(P 〉 0.05). (2) The positive rate of p53 protein was 67.6 % and significantly higher than that in normal epithelium ( P〈 0.05). The expression of p53 protein in the clinical stage Ill, 1V were significantly higher than that in clin- ical stage Ⅰ , Ⅱ (P 〈 0.05). (3)There was correlation between the expression of β- catenin and the expression of p53 protein(r = 0. 373, P〈 0.05). Conclusion Abnormal expression of β - catenin and overexpression of p53 protein may be individually or conjunctly involved in the course of pathogenesis and development of oral squa- mous cell carcinoma.
