摘要
【目的】建立适合五种南疆特色果树核桃、香梨、苹果、杏、扁桃的高质量RNA提取方法。【方法】采用试剂盒法、传统CTAB法、改良硼砂-CTAB法和改良硼砂-CTAB-异丙醇法等四种方法提取核桃花芽、香梨花芽、苹果花芽、杏叶片以及扁桃叶片总RNA,并比较提取效果。【结果】试验方案2(萌芽期N+P2O5+K2O19.2 kg/667m2+19.2 kg/667m2+11.6 kg/667m2,开花期N+P2O5+K2O 8 kg/667m2+12 kg/667m2+10 kg/667m2,坐果期N+P2O5+K2O 6.7 kg/667m2+6.7 kg/667m2+16.6 kg/667m2)比较适合幼龄枣树的施肥。【结论】与CK及其它施肥方案相比,在施肥方案2下,幼龄枣树株高、二次枝粗度、主干干径、新梢增长量、叶片面积、叶绿素含量以及叶片氮、磷、钾的累积吸收较佳,产量较高。
【Objective】The study aims to establish several RNA extraction methods which might be of high quality and suitability for Southern Xinjiang five featured fruit trees in the hope of paving the way for subsequent research.【Method】The kit method,traditional CTAB,modified borax-CTAB and improved borax-CTAB-isopropanol RNA isolation methods were used to isolate total RNA from walnut buds,pear buds,apple buds,apricot leaves and almond leaves,and the corresponding results were compared in this study.【Result】Fertilization experiment scheme 2( Formingstage: N + P2O5+ K2 O 19. 2 kg /667m^2+ 19. 2 kg /667m^2+11. 6 kg /667m^2,Blossom period: N + P2O5+ K2 O 8 kg /667m^2+ 12 kg /667m^2+ 10 kg /667m^2,Fruiting period: N + P2O5+ K2O6. 7 kg /667m^2+ 6. 7 kg /667m^2+ 16. 6 kg /667m^2) is more suitable for young jujube.【Conclusion】The indexes of plant height,secondary branch coarse,trunk diameter,shoot increment,blade area,chlorophyll content,accumulation and absorption of N,P,K of young jujube are better than CK,and had a higher yield,under the effect of fertilization program 2.
