摘要 【目的】研究抗病性影响因子连锁的分子标记与抗黄萎病性状的一致性,筛选抗黄萎病基因,发掘其与棉花抗黄萎病性状的关系,进一步阐明棉花抗黄萎病遗传机制。【方法】研究使用37对抗黄萎病性影响因子设计的引物进行PCR扩增来探明抗病性基因在不同抗、感病棉花品种中的分布,以及与抗黄萎病性状的一致性。【结果】发现VM13、VM23、VM25、BNL0946、BNL1414、BNL1721、em6me2和em1me5这8对引物扩增出不同条带,分别来源于抗病相关蛋白的基因、基因组序列、mRNA序列、SSR和SRAP引物;其中,引物VM13、BNL1414、em6me2扩增条带与抗病品种一致率高于0.857,与感病品种一致率低于0.25。新疆自育品种较多出现的扩增条带,与新疆自育品种一致率介于0.364-0.727,平均值为0.515;与其它地区所育品种的一致率介于0.111-0.666,平均值为0.419。【结论】引物VM13、BNL1414、em6me2的扩增条带可以作为黄萎病抗性的分子标记鉴定指标。 【Objective】The purpose of this study is to search the consistency of molecular markers linked with factors affected Verticillium wilt resistance and resistant levels in cotton in order to get the further study about the wilt resistant genes and genetic mechanism of resistance. 【Method】Thirty-seven pairs of primers derived from Verticillium w. resistance factors were used in PCR to investigate the distribution of wilt resistant genes among resistant,tolerant,and susceptible cotton cultivars,and the consistency of selected markers with the resistant performance of cotton cultivars. 【Result】The results indicated that eight makers VM13、VM23、VM25、BNL0946、BNL1414、BNL1721、em6me2 and em1me5 produced polymorphic fragments. These primers originated from either disease-resistant protein、orgenomic、mRNA sequence、SSR and SRAP that were associated with Verticillium w. resistance. The consistency index of the PCR fragments appearing in the majority of resistant cultivars for the primers VM13、BNL1414 and em6me2 within resistant cultivars were grea-ter than 0. 857,while within susceptible cultivars were lower than 0. 25. The consistency index of the PCR fragments appearing in the majority of Xinjiang bred cultivars within Xinjiang bred cultivars ranged 0. 364 -0. 727 with an average of 0. 515,while in other region bred cultivars ranged 0. 111 - 0. 666 with an average of0. 419. 【Conclusion】The polymorphic bands amplified by the three,VM13、BNL1414 and em6me2,serve as good molecular markers for Verticillium w. resistance in cotton.
