狼毒大戟二萜类成分HPLC指纹图谱及聚类分析 被引量:7

摘要 为建立狼毒大戟药材的特征图谱,科学评价其质量,本文采用高效液相色谱法,乙腈-水梯度洗脱,测定了11批不同来源的狼毒大戟样品二萜类化合物,通过相似度分析建立了狼毒大戟提取物的高效液相指纹图谱,并对11批药材进行系统聚类分析。本文采用乙腈-水为流动相,梯度洗脱,检测波长:230 nm,流速:1.0mL/min,进样量:10μL,柱温:30℃。相似度评价结果显示11批不同产地的狼毒大戟相似度均高于0.9,并对11批样品建立共有模式,确定了23个特征共有峰,聚类分析结果将狼毒大戟样品分为2类,与药材产地相关。本文所建立狼毒大戟的指纹图谱方法简便、可靠,可作为狼毒大戟药材的基源鉴定参考方法。 This study was aimed to establish the method of fingerprint analysis of chemical constituents of Euphorbiafis- cher/ana by reverse-phase high-performance liquid chromatography (HPLC) and hence to control its quality. A mixed mobile phase of water and aeetonitrile in a gradient elution mode was applied to determine diterpenoid compounds in 11 batches of E.fischer/ana. The flow rate was 1.0 mL/min;the UV detection wavelength was 230 nm;the injection volume was 10 μL;the column temperature was 30 ℃. In addition, classification of different E.fischeriana samples was obtained using systematic duster method. The analysis results of Similarity Evaluation System for Chromatographic Fingerprint suggested that the degree of similarity of the 11 batches of E. fischeriana samples were over 0.9. The HPLC chromatographic fingerprint of chemical constituents in the 11 batches of E.fischer/ana was established, which showed 23 characteristic common peaks. The cluster analysis results indicated that the 11 batches of samples were classified into 2 classes ,which was related to the origins of herbs. The developed HPLC method can be used to identify the origins of E.fischerrana herbs and to control its quality.
