摘要
目的:探讨消瘤汤对结直肠癌移植瘤裸鼠血清VEGF-A、VEGF-C蛋白表达的影响。方法:SPF级裸小鼠60只按随机数字表分为治疗组(消瘤汤正常剂量组、贝伐单抗组及消瘤汤高、低剂量组)和对照组,取结直肠癌患者新鲜手术标本切成小植块移植到裸鼠皮下,肿瘤成长后再分别取小植块经手术移植到一批裸鼠结肠系膜侧。移植后,消瘤汤正常剂量组给予消瘤汤正常剂量灌胃;贝伐单抗组给予贝伐单抗静脉注射;消瘤汤高、低剂量组分别予消瘤汤高、低剂量灌胃;对照组给予蒸馏水灌胃。观察治疗组和对照组裸鼠血清VEGF-A、VEGF-C蛋白表达情况。结果:各治疗组裸鼠血清VEGF-A、VEGF-C蛋白表达水平明显低于对照组,差异有统计学意义(P<0.05);消瘤汤组与贝伐单抗组及消瘤汤高、低剂量组比较,差异均无统计学意义(P>0.05)。结论:消瘤汤能抑制结直肠癌患者血清VEGF-A、VEGF-C蛋白表达可能是消瘤汤抗肿瘤的作用机制之一。
Objective:To explore the effects of Xiaoliu Decoction on the expressions of vascular endothelial growth factor A(VEGF-A) and vascular endothelial growth factor C(VEGF-C) in serum of nude mice with colorectal transplantation tumor.Methods:60 nude mice of SPF were divided into the treatment group(Xiaoliu Decoction normal dose group,bevacizumab group,Xiaoliu Decoction high dose group and Xiaoliu Decoction low dose group) and the control group by the random numbers.Colorectal cancer patients' fresh surgical specimens were cut into small pieces and transplanted into subcutaneous of nude mice.After tumor growth,we took out small plant blocks to transplant to mesangium side of colon of nude mice by operation.The Xiaoliu Decoction normal dose group used oral Xiaoliu Decoction normal dose by irrigation stomach.The bevacizumab group used intravenous injection of bevacizumab.The Xiaoliu Decoction high dose group and low dose group respectively used oral Xiaoliu Decoction high dose and low dose by irrigation stomach.The control group used oral distilled water by irrigation stomach.We observed treatment group and the control group's expressions of VEGF-A and VEGF-C in serum of nude mice with colorectal transplantation tumor.Results:The expressions of VEGF-A and VEGF-C in serum of nude mice of the treatment group were obviously lower than that of the control group(P〈0.05).The Xiaoliu Decoction normal dose compared with the bevacizumab group,Xiaoliu Decoction high dose group and Xiaoliu Decoction low dose group had no statistically significant difference(P〉0.05).Conclusion:Xiaoliu Decoction can inhibit the expressions of VEGF-A and VEGF-C in serum of patients with colorectal cancer,which is possibly one of the mechanisms of Xiaoliu Decoction in anti-tumor.
