骨碎补总黄酮对牙周炎大鼠牙周组织中白细胞介素-6和骨钙素表达的影响 被引量:5

摘要 目的观察骨碎补总黄酮对牙周炎大鼠牙周组织中白细胞介素-6(IL-6)和骨钙素(BGP)水平的影响,探讨其治疗牙周炎的机制。方法实验采用SPF级雄性SD大鼠54只,随机分为3组:正常组,牙周炎组(参照吴亚菲等的方法复制大鼠实验性牙周炎模型),骨碎补总黄酮组(复制大鼠实验性牙周炎模型,并于建模后第5周开始用骨碎补总黄酮灌胃)。各组大鼠分别在建模后第4,6,8周分批处死,各时点每组处死动物数为6只。免疫组织化学方法检测牙周组织中IL-6、BGP的表达。结果牙周炎组牙周组织IL-6水平在6周和8周时显著高于正常组(P<0.05);骨碎补总黄酮组牙周组织IL-6表达水平在6周和8周时显著低于牙周炎组(P<0.05),并与正常组比较差异无统计学意义(P>0.05)。牙周炎组牙周组织BGP水平在6周和8周时显著低于正常组(P<0.05);骨碎补总黄酮组牙周组织BGP表达水平在6和8周时显著高于牙周炎组(P<0.05)。结论骨碎补总黄酮促进牙周炎大鼠牙周组织BGP的表达,抑制IL-6的表达,对牙周炎的治疗具有重要意义。 Objective To explore the effect of rhizoma drynariae flavone on levels of cytokine interleukin-6(IL-6) and bone gla protein (BGP) in the periodontal tissues of rat periodontitis model and its mechanism. Methods Fifty-four SD male rats (SPF grade)were randomly divided into three groups: normal group, periodontitis group, and periodontitis + rhizoma drynariae flavone group. The experi- mental periodontitis model was established according to the method reported by Wu Yafei. In rhizoma drynariae flavone + periodontitis group, the periodontitis model rats were fed with rhizoma drynariae flavone at week 5 after modeling. Six rats were were executed at week 4, 6, 8 in each group after modeling by batch, respectively. The levels of IL-6 and BGP in the periodontium were measured by immunohistochemistry. Results The level of IL-6 in periodontitis group was significantly higher than that in normal group at week 6 and 8 ( P 〈 0.05 ). The level of IL-6 in periodontitis + rhizoma drynariae flavone group was significantly lower than that in periodontitis group at week 6 and 8 ( P 〈 0. 05 ), and there was no statistical difference between periodontitis + rhizoma drynariae flavone group and normal group(P 〉 0.05 ). The level of BGP in periodontitis group was significantly lower than that in normal group at week 6 and 8 ( P 〈 0.05 ), and the level of BGP in periodontitis + rhizoma drynariae flavone group was significantly higher than that in periodontitis group at week 6 and 8 (P 〈0. 05). Conclusion Rhizoma drynariae flavone could play an important role in treating periodontitis by promoting the expression of BGP in periodontal tissues of rat periodontitis model and inhibiting the expression of IL-6.
