三苯双脒治疗旋毛虫感染小鼠血清IL-4和TGF-β1的变化 被引量:3

Changes in IL-4 and TGF-β1 in mice infected with Trichinella spiralis after tribendimidine treatment

摘要 目的观察旋毛虫感染小鼠经三苯双脒治疗后血清中细胞因子IL-4和TGF-β1水平变化。方法将120只昆明小鼠随机分成,感染对照组、治疗组和正常对照组,每组40只。感染对照组和治疗组小鼠经灌胃感染云南株旋毛虫肌幼虫200条/只。治疗组小鼠于感染后第7d经灌胃给予三苯双脒治疗,每只每天300mg体重,连续7d。分别在感染后7、14、21、28和35d对各组小鼠采血,分离血清,采用ELISA检测IL-4和TGF-β1水平。结果 ELISA检测显示,感染对照组和治疗组IL-4、TGF-β1水平在感染旋毛虫后开始升高,在感染后14d达到峰值,分别为(40.17±1.01)pg/ml、(100.23±1.31)pg/ml、和(30.21±1.25)pg/ml、(80.51±1.37)pg/ml,随后逐渐下降。在感染后5个时间点,感染对照组小鼠血清IL-4和TGF-β1水平与正常对照组比较差异均有统计学意义(P<0.01);治疗组IL-4和TGF-β1水平在感染后5个时间点与感染对照组(P<0.01),与正常对照组比较差异均有统计学意义。结论三苯双脒治疗旋毛虫感染小鼠后血清IL-4和TGF-β1水平随感染时间先升高后降低。 Objective To observe changes in IL-4and TGF-β1in mice infected with Trichinella spiralis after tribendimidine treatment. Methods One hundred and twenty Kunming mice were randomly divided into an infection control group,a treatment group,and a normal control group with 40 mice each.Mice in the infection control group and treatment group were each fed 200 muscle larvae of the Yunnan strain of T.spiralis.Seven days after infection,mice in the treatment group were orally administered tribendimidine at a dose of 300mg/(kg·d)for 7days straight.Blood was collected 7,14,21,28,and 35 days after treatment.Serum was separated and IL-4and TGF-β1levels were measured using ELISA. Results ELISA indicated that IL-4and TGF-β1levels in the infection control group and treatment group began to increase after infection and peaked 14 days after infection(40.17±1.01pg/ml and 100.23±1.31pg/ml for the infection control group and 30.21±1.25pg/ml and 80.51±1.37pg/ml for the treatment group).Afterwards,those levels gradually decline.At five time points after infection,serum levels of IL-4and TGF-β1in the control group differed significantly(P〈0.01)from those in the normal control group.Levels of IL-4and TGF-β1differed significantly in the infection control group(P〈0.01)and the normal group(P〈0.05). Conclusion After tribendimidine treatment,the serum levels of IL-4and TGF-β1in infected mice initially rose and then declined.
