软坚消瘿颗粒对AIT大鼠细胞因子Th1/Th2及凋亡蛋白Fas/FasL、Bcl-2/Bax的影响 被引量:13

摘要 目的通过对AIT大鼠软坚消瘿汤改变剂型后颗粒剂的疗效,观察细胞因子(Th1/Th2)及甲状腺细胞凋亡相关调控蛋白Fas/Fas L、Bcl-2/Bax的影响,并探讨其作用机制。方法将48只雌性大鼠按体重大小随机分为4组,即正常组、模型组、雷公藤组、软坚消瘿颗粒组。造模采用腹部皮下多点免疫注射猪甲状腺球蛋白与弗氏佐剂溶合物,同时配合饮用高碘水制备AIT大鼠模型。经过6 w治疗后,检测以下指标:采用免疫组化法测定大鼠细胞因子Th1,Th2及甲状腺组织凋亡蛋白Fas、Fas-L、Bax、Bcl-2的表达;同时测定以上4种凋亡蛋白的灰度值。结果 (1)凋亡蛋白的变化:模型组与正常组比较Fas、Fas-L、Bax显著升高,Bcl-2显著降低(P<0.01);疗后软坚消瘿颗粒组及雷公藤组与模型组比较有显著差异(P<0.01);治疗二组比较无显著性差异(P>0.05);(2)细胞因子Th1/Th2的变化:模型组与正常组比较Th1/Th2显著升高(P<0.01);疗后软坚消瘿颗粒组及雷公藤组与模型组比较有显著差异(P<0.01);治疗二组比较无显著性差异(P>0.05)。结论 (1)软坚消瘿颗粒组及雷公藤组可有效降低细胞因子Th1/Th2,从而起到治疗作用;(2)软坚消瘿颗粒组及雷公藤组通过调控凋亡蛋白,起到抑制细胞凋亡的作用,从而减轻或延缓本病的发生与发展。 Objective To observe the effect of Ruanjianxiaoying Granule on cytokines( Th1 / Th2) of AIT rats and apoptosis proteins Fas / Fas L,Bcl- 2 / Bax,exploring the mechanism. Methods 48 female rats were randomly divided into four groups,namely the normal group,model group,triptolide group,Ruanjianxiaoying granule groups. The model of rats was established by injecting the mixture of pig thyroglobulin and Freund' s adjuvant subcutaneously at various points of abdomen,with more high iodine water taken in. After six weeks' Ruanjianxiaoying treatment,the following indicators were tested: Th1 / Th2 and the expression of apoptosis proteins Fas,Fas L,Bax and Bcl- 2 were determined by immunohistochemical method,and the grey value of above proteins were determined at the same time. Results( 1) apoptosis proteins: compared with the normal group,Fas,Fas- L,Bax of the model group were distinctly higher,while Bcl- 2 was much lower( P〈0. 01),those of Ruanjianxiaoying granule group and tripterygium group were obviously different from the model group( P〈0. 01),there were no significant differences between the two treated groups( P groups 〉0. 05).( 2) Th1 / Th2: those of the model group were much higher than those of the normal group( P〈0. 01),those of Ruanjianxiaoying granule group and tripterygium group were obviously different from the model group( P〈0. 01),the treatment of the two groups showed no significant difference( P〉0. 05). there were no significant differences between the two treated groups( P groups〉 0. 05). Conclusion( 1) Ruanjianxiaoying granule and tripterygiumcan effectively reduce cytokine Th1 / Th2,thus can serve as a therapy.( 2) Ruanjianxiaoying granule and tripterygium can inhibit apoptosis of cells through regulating apoptosis proteins,alleviating or delaying the occurrence and development of the disease.
