摘要
以"通蓖5号"单粒蓖麻种子为研究对象,利用BCA试剂盒、UVwin5紫外软件V5.1.0、Band Scan蛋白定量分析软件相结合的方法,优化了单粒蓖麻种子的粗蛋白提取过程,并对单粒蓖麻种子中毒蛋白含量进行测定。结果如下,优化后的单粒蓖麻种子粗蛋白提取过程中缓冲液A的加入量为3m L/g,硫酸铵饱和度为70%,沉淀时间是12h,溶解沉淀所需加入缓冲液A为3m L。经测定,单粒蓖麻种子平均质量为0.2415g,其中含粗蛋白10.8908mg,毒蛋白4.1154mg,毒蛋白所占百分比为37.79%。
The single "Tongbi5" castor seed was the study object in this paper.Aiming for optimizing extraction pro-cess of crude protein,a variety of methods of which BCA kit,UVwin5 ultraviolet software(v5.1.0) and Band Scan protein quantitative analysis software were used.The results were as follows,in the optimized extraction process of crude protein of a castor seed,the amount of buffer A is 3m L/g,time of precipitation with ammonium sulfate(70% satu-ration) is 12 h,and the amount of buffer A to dissolve precipitation is 3m L.It was found by measurement that the average quality of a castor seed is 0.2415 g,the average content of crude protein is 10.8908 mg.Ricin makes up crude protein by 37.79 and content of ricin is 4.1154 mg on average in a castor seed.
