疏风清肺化痰汤对慢性支气管炎模型大鼠的保护作用研究

Study on the Protective Effect of Shufeng Qingfei Huatan Decoction on Model Rats with Chronic Bronchitis

目的:研究疏风清肺化痰汤对慢性支气管炎模型大鼠的保护作用。方法:实验分为正常对照(0.9%氯化钠溶液,10 ml/kg)组、模型(0.9%氯化钠溶液,10 ml/kg)组、红霉素(0.14 g/kg)组与疏风清肺化痰汤高、中、低剂量[4.32、2.16、1.08 g(生药)/kg]组,后5组以改良烟熏法复制大鼠慢性支气管炎模型。复制模型成功后ig给药,连续28 d。测定大鼠右肺组织匀浆中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)的活性和丙二醛(MDA)的含量。取大鼠左肺组织进行HE切片,显微镜下观察病理变化。结果:与正常对照组比较,模型组大鼠肺组织匀浆中SOD、GSH-Px活性减弱,MDA含量增加,差异有统计学意义(P<0.01);病理观察呈重度炎症表现。与模型组比较,疏风清肺化痰汤高、中、低剂量组肺组织匀浆中SOD、GSH-Px活性增强,MDA含量减少,差异有统计学意义(P<0.01或P<0.05);病理观察均见一定程度减轻。各组CAT比较差异均无统计学意义(P>0.05)。结论:疏风清肺化痰汤对慢性支气管炎模型大鼠具有一定的保护作用。

OBJECTIVE ： To study the protective effect of Shufeng qingfei huatan decoction on model rats with chronic bronchi- tis. METHODS： There were normal control group （0.9% sodium chloride solution, 10 ml/kg）, model group （0.9% sodium chlo- ride solution, 10 ml/kg）, erythrocin group （0.14 g/kg） and Shufeng qingfei huatan decoction high, medium and low dose groups [4.32, 2.16, 1.08 g（medicinal materials）/kg]. Rat models of chronic bronchitis were established for latter 5 groups by modified smoking method. After successful establishment of models, the rats were given drugs for 28 days, ig. The activities of superoxide dismutase （SOD）, catalase （CAT） and glutathione peroxidase （GSH-Px） and the content of malondialdehyde （MDA） in the ho- mogenate of right lung tissues of rats were determined. The left lung tissues were collected to prepare the sections for HE staining, and the pathological changes were observed under the microscope. RESULTS： Compared with normal control group, the activities of SOD and GSH-Px in model group were decreased and the content of MDA was increased, with significant difference （P〈 0.01） ; there was serious inflammation in the pathological observation. Compared with model group, the activities of SOD and GSH-Px in Shufeng qingfei huatan decoction high, medium and low dose groups were increased and MDA was decreased, with sig- nificant difference （P〈0.01 or P〈0.05） ; there was less serious inflammation. There were no significant differences in the CAT among all groups （P〉0.05）. CONCLUSIONS： Shufeng qingfei huatan decoction has certain protective effect on model rats with chronic bronchitis.
