摘要
旨在研究自拟中草药添加剂对绵羊肉中鲜味物质质量分数和腺苷琥珀酸裂解酶基因(ADSL)表达的影响。32只绵羊随机分为4组,空白组仅饲喂基础日粮,试验组按照日粮质量分数1%的剂量分别添加中草药方剂Ⅰ、方剂Ⅱ和方剂Ⅲ,饲喂60d后,采用高效液相色谱(HPLC)法检测肉样中肌苷酸(IMP)及其相关代谢物的质量分数,实时荧光定量PCR(FQ-PCR)法检测ADSL基因相对表达水平,分析ADSL基因表达与IMP质量分数的相关性。结果表明,所有试验样品中均不含三磷酸腺苷(ATP)成分,二磷酸腺苷(ADP)和一磷酸腺苷(AMP)的质量分数亦很低,各组间差异不显著(P>0.05);3个中草药方剂均能不同程度地提高绵羊肉中IMP及其分解产物的质量分数,其中方剂Ⅲ组的IMP、肌苷(INO)和校正肌苷酸(IMPc)值极显著地高于对照组(P<0.01),与方剂Ⅰ、Ⅱ组相比差异显著(P<0.05);方剂Ⅲ组羊肉样中ADSL基因相对表达水平与空白组相比存在显著性差异,且ADSL基因表达与IMP、IMPc呈显著正相关(P<0.05)。说明,中草药方剂Ⅲ可显著改善绵羊肉风味,ADSL基因可作为肉质风味的候选基因。
The experiment was conducted to investigate the effects of Chinese herb feed additives on the umami substances mass fraction and ADSLrelative expression level of mutton.32 healthy 10-month-old Chinese Merino sheep were randomly divided into 4groups.The control group was given basal diet.The treatment groups were fed basal diets added with 0.1% Chinese herb feed additiveⅠ,0.1% Chinese herb feed additiveⅡ and 0.1% Chinese herb feed additive Ⅲ,respectively.After 60 days of feeding,the mass fractions of inosinic acid(IMP)and its correlative metabolites of samples were determined by HPLC method,and ADSLrelative expression level was detected by real-time fluorescent quantitative PCR(FQ-PCR);meanwhile,the correlation between ADSLexpression and umami substances mass fraction was analyzed.The results showed that ATP was not detected in all of the samples;the mass fraction of ADP and AMP were very low,and there was no significant intergroup difference(P〉0.05);three Chinese herb feed additives all can improved the mass fraction of IMP andIts correlative metabolites in different degrees;the IMP,INO and IMPc mass fraction of Chinese herbⅢ were extremely significantly higher than the control group(P〈0.01),and significantly higher than the Chinese herb Ⅰ and Chinese herb Ⅱ(P〈0.05);ADSLgene expression level of Chinese herb Ⅲgroup was significantly different compared with the control group;there was significant positive correlation between ADSLgene expression and IMP as well as IMPc.The results demonstrated that the Chinese herb feed additiveⅢ can significantly improve the flavor of mutton,and ADSLgene can be used as flavour candidate gene for further research.
