摘要
采用ISSR分子标记技术,对异裂菊属植物进行亲缘关系分析。从100条引物中筛选得到13条多态性较好的引物。ISSR反应共得到106条条带,多态性条带有84条,多态位点百分率为78.98%。种间遗传距离的变化范围在0.400 8-0.620 4之间,遗传一致度的变化范围在0.537 7-0.669 8之间。异裂菊属种间相似性的UPGMA聚类结果表明,凹脉异裂菊和小花异裂菊亲缘关系较近,异裂菊和柳州异裂菊亲缘关系较近,绢叶异裂菊与其他4个种的关系较远。ISSR表型特征的主成分分析也支持聚类分析结果。
The genetic relationship of Heteroplex Chang were ananlysed by ISSR. 13 primers were selected out from 100 random primers which can yield specific bands with moderate band numbers. Total 106 specific and reproducible bands were obtained of which 84 were polymorphic bands which polymorphism were 78. 98%.Genetic identity and genetic distance showed that variance ranged from 0. 537 7 to 0. 669 8,and 0. 400 to0. 620 4. Cluster ananlysis of the Jaccard coefficient matrix using UPGMA method revealed that genetic relationship were low between Heteroplexis microcephala Y. L. chen and Heteroplexis impressinervia J. Y. Liang,Heteroplexis vernonioides Chang and Heteroplexis incana J. Y. Liang,relationship was high between Heteroplexis sericophylla Chang and other four species. which also lent support from the spatial representation of principle component analysis of ISSR phonetic patterns.
