摘要
目的观察丹参酮ⅡA对压力负荷增加大鼠心室胶原重构的影响。方法采用腹主动脉缩窄术制备压力负荷增加大鼠模型,动脉夹闭(其中8只大鼠不结扎腹主动脉,为假手术组)4周后,分为5组:假手术组、模型组、丹参酮ⅡA低剂量组[10 mg/(kg·d)]、丹参酮ⅡA高剂量组[20 mg/(kg·d)]及卡托普利组[100 mg/(kg·d)]组,每组各8只,各组均予相应药液腹腔注射(卡托普利组为灌胃给药),连续给药4周后检测各组大鼠心脏质量指数(HWI)和左心室质量指数(LVWI),苏木精—伊红(HE)染色法观察左心室心肌病理学改变,Masson染色法观察左心室心肌胶原形态,化学比色法测定心肌羟脯氨酸(HYP)含量。结果与假手术组比较,模型组大鼠HWI、LVWI、心肌组织HYP含量均明显升高(P<0.05,P<0.01),心肌有不同程度的损害;与模型组比较,卡托普利组、丹参酮ⅡA高剂量组及丹参酮ⅡA低剂量组HWI、LVWI及HYP均明显下降(P<0.01,P<0.05),心肌损害均有不同程度改善。结论丹参酮ⅡA能够抑制压力负荷增加大鼠心肌胶原沉积和心肌HYP的表达,在一定程度可改善心室胶原重构。
Objective To observe the effects of tanshinoneⅡA ( TanⅡA) on ventricular collagen remode-ling in rats with pressure overload increased.Methods The myocardial fibrosis rats with pressure overload increased were prepared with abdominal aortic constriction.These rats received abdominal aorta ligation for 4 weeks ( 8 rats of them didn&#39;t ligate abdominal aorta, as sham operation group) .The rats were divided into five groups:sham operation group, model group, TanⅡA of low dose group [ L-TanⅡA group,10 mg/( kg·d) ] , TanⅡA of high dose group [H-TanⅡA group, 20 mg/(kg·d)] and captopril group[100 mg/(kg·d)],8 rats in each group.The corre-sponding medicine in each group was given by intraperitoneal injection( captopril group was given by intragastric ad-ministration) , successive administration 4 weeks.After 8 weeks of surgery, heart mass index ( HMI) and left ventric-ular mass index ( LVMI) were detected, left ventricular myocardial histology were observed by HE staining, as well as left ventricular myocardial collagen forms by Masson staining, and ELISA for myocardial hydroxyproline ( HYP) content.Results HMI, LVMI and myocardial HYP content in model group were significantly higher than those in sham operation group(P〈0.05,P〈0.01), and there were myocardial damage at different degree.Compared with model group, HMI, LVMI and myocardial HYP content in other three groups were obviously decreased (P〈0.05,P〈0.01), and there were improvement of myocardial damage at different degree.Conclusion TanshinoneⅡA can markedly inhibit myocardial collagen deposition and myocardial expression of HYP in pressure overload increased rats, and improve ventricular collagen remodeling in some extent.
