摘要
以甘油为原料,通过单因素实验对菌株间歇发酵法生产1,3-丙二醇的生产条件进行了研究.结果表明:最适的甘油含量为40g/L,最适氮源为(NH4)2HPO4,用量为3g/L,pH 7.5,温度为30℃,装液量为40%,摇床转速为100r/min,发酵时间为24h.在最佳培养基组成与最佳发酵条件下发酵了3批,最终的1,3-PDO浓度平均值为23.2g/L,甘油的摩尔转化率为70.2%,比优化前的60.5%提高了9.7%.
Through single-factor test,conditions of disproportionation fermentation by glycerol for 1,3-propanediol(PDO)by bacteria were optimized.The results show that the optimal glycerol content is40g/L,the optimum nitrogen source is(NH4)2HPO4 with dosage of 3g/L,pH of 7.5,temperature of 30℃,liquid volume of 40%,shaking speed of 100r/min,and fermentation time of 24 h.In the optimal medium and fermentation conditions,three batches of experiments were carried out and the average concentration of 1,3-PDO could reach 23.2g/L,which raised the molar yield of glycerol from 60.5%to 70.2%.
