摘要
采用沃尔巴克氏体(Wolbachia)的通用引物、A大组和B大组的特异性引物对烟蚜茧蜂体内Wolbachia的wsp基因进行PCR扩增及分子检测,所获得的基因片段分别命名为w Apr、w Apr A和w Apr B,长度分别为620 bp、566 bp和463 bp。基因序列分析表明,w Apr、w Apr A和w Apr B在对应位置序列仅有4个碱基的差异。系统发育分析表明,该寄生蜂仅感染了A大组的Wolbachia。
PCR amplification was employed to detect the infection of Wolbachia in Aphidius gifuensis by using universal primers and specific primers for wsp gene of A and B supergroups. Sequences obtained were named as wApr, w Apr A and w Apr B with sizes of 620 bp, 566 bp and 463 bp, respectively. Sequence analysis indicated that 4 variable base pairs occurred among these sequences. Phylogenetic tree analysis of wsp gene showed that Wolbachia in A. gifuensis were classified as A group.
