摘要
采用碱-还原两浴两步法溶解人发,提取角蛋白。实验得出的最佳工艺为:0.1 mol/L Na OH处理1 h,采用还原体系(0.75 mol/L Na2SO3,8 mol/L尿素,0.02 mol/L SDS),在80℃水浴中溶解5 h,浴比1∶15,人发溶解率可达55%;对提取的角蛋白进行SDS-PAGE测试及FTIR测试,结果表明:角蛋白的分子量主要集中在25~37 k Da,其结构以α-螺旋为主,同时含有β-转角、β-折叠及无规则卷曲。
A new method of alkali-reduction with two-bath in two-step was used to extract keratin from human hair efficiently. The optimum condition was concluded, a mass of human hair was processed with 0. 1 mol/L NaOH for 1 h,then was mixed with 0.75 mol/L NaSO3 ,8 mol/L urea,0.02 mol/L SDS according to a ratio of 1:15 ,and was deoxygenized at 80℃ for 5 h,its dissolution rate was around 55%. The keratin was analysed by SDS-PAGE and FTIR. The result revealed that the mass of the keratin is centralized between 25 kDa and 37 kDa. The extracted keratin was mainly composed by random-coil and conserved eL-helix, 13-Turn, 13-sheet.
