槲皮素对小细胞肺癌细胞株H446的凋亡调控作用 被引量:5

摘要 目的初步探讨槲皮素对人小细胞肺癌细胞株凋亡的影响。方法以不同浓度槲皮素培养H446细胞24 h后,运用免疫荧光染色法检测细胞中抗凋亡蛋白Bcl-2的表达情况,同时采用流式细胞仪检测细胞凋亡率的变化。结果以0、15、30、60μmol/L浓度的槲皮素作用于细胞株24 h后,细胞凋亡率分别为(1.30±0.32)%(12.95±0.08)%,(16.8±0.19)%,(25.81±1.01)%,各组间均值具有显著性差异(P<0.01),并且随着槲皮素浓度的增加,H446细胞凋亡率逐渐增加,两者呈正性相关。采用免疫荧光染色法检测发现,随着槲皮素作用浓度的增加,抗凋亡蛋白BCL-2的表达信号亦逐渐降低。结论槲皮素对人小细胞肺癌细胞株有促进凋亡的作用,其机制可能与抑制抗凋亡蛋白BCL-2的表达有关。 Objective To study the influence of apoptosis induced by quercetin on small-cell lung cancer cells. Methods H446 cells were cultured with quercetin at different doses for 24h. Immunofluorescence was performed to detect the expression of BCL-2 in H446 cells, and the flow cytometry was used to test the apoptosis rate of H446 cells incubated with quercetin of different concentration. Results The apoptosis rate was (1. 30 ± 0. 32)%, (12. 95 ± 0. 08)%, (16. 8 ± 0. 19)%, and (25. 81 ± 1. 01)% respectively at the dose of 0, 15, 30 and 60 umol/L (P〈0. 05), showing a positive correlation with the dose increase. The expression of Bcl-2 in H446 cells was inhibited by quercetin. Conclusion The apoptosis induced by quercetin in small cell lung cancer cells may be related with the reduction in expression of protein BCL-2.
