白藜芦醇对缺氧缺血性脑损伤新生大鼠MMP-9及TIMP-1的影响 被引量:3

摘要 目的探讨白藜芦醇对缺氧缺血性脑损伤新生大鼠MMP-9及TIMP-1的影响。方法选择健康7日龄新生Wistar大鼠112只,分为三组:假手术组(n=36只)、缺氧组(n=38只)、HIBD+白藜芦醇药物干预组(n=38只),各组再分为6 h、24 h、48 h、72 h、5 d和7 d时间段进行检测。比较各组不同时间点MMP-9、TIMP-1的表达。结果HIBD组的MMP-9水平治疗6 h、24 h、48 h、72 h、5 d、7 d分别显著高于假手术组,但经RES干预后,HIBD+RES组的MMP-9水平分别显著低于HIBD组(P<0.05),但仍显著高于假手术组。结论 RES能够抑制MMP-9的表达,从而使伴随MMP-9病理性表达升高的TIMP-1也相应减少,而调节MMP-9/TIMP-1的平衡,可能是其在HIBD中发挥神经保护作用的机制之一。 Objective To investigate the impact of resveratrol on MMP-9 and TIMP-I for hypoxic ischemic brain dam- age in neonatal rats. Methods Selected healthy 7 day old newborn Wistar rats 112 rats,were divided into three groups: sham operation group (n=36), hypoxia group (n=38 only), HIBD+resveratrol drug intervention group (n=38), each group was tested at 6 h, 24 h, 48 h, 72 h, 5 d and 7 d. The expression of TIMP-1 and MMP-9 Groups were com pared at different time points. Results The MMP-9 level of 6 h, 24 h, 48 h,72 h, 5 d, 7 d in HIBD group, were sig- nificantly higher than those in the sham operation group,and was significantly higher than that of RES, but the MMP- 9 levels in intervention group HIBD+RES were significantly lower than those in HIBD group(P〈0.05), but still signifi- cantly higher than those in the shamoperation group. Conclusion The expression of RES can inhibit MMP-9, so with MMP-9 diseaseand rational expression of elevated TIMP-1 also reduce, and regulate the balance of MMP-9/TIMP-1, may be the one of the nerve protection mechanism in HIRD.
