靶向VEGF-C的siRNA表达载体对人乳腺癌细胞VEGF-C基因表达的影响 被引量:2

摘要 目的探讨靶向VEGF-C的siRNA表达载体对人乳腺癌细胞MCF7增殖、凋亡及化疗敏感性的影响。方法针对VEGF-C的siRNA表达载体转染乳腺癌细胞,另设立转染组及转染阴性表达载体组,每组设5个复孔。观察各组细胞的VEGF-C mRNA及蛋白表达情况。结果阴性对照组和空白对照组VEGF-C mRNA表达率无显著差异(P>0.05);实验组各组的表达水平均显著低于空白对照组及阴性对照组(P<0.01);实验A组的表达又显著低于实验B组和实验C组(P<0.01)。阴性对照组和空白对照组培养液上清中VEGF-C蛋白水平比较,差异无统计学意义(P>0.05);实验组各组蛋白水平均显著低于阴性对照组和空白对照组(P<0.01);实验A组蛋白水平又显著低于实验B组和实验C组(P<0.01)。结论靶向VEGF-C的siRNA表达载体能够显著降低人乳腺癌细胞VEGFC的基因和蛋白表达。 Objective To discuss the effect of siRNA expression vector of targeting VEGF-C on breast cancer cells VEGF-C gene expression. Methods The siRNA expression vector of targeting VEGF-C transfected breast cancer cells, and another groups of expression vector transfected and transfected with negative were established. Each group set up five wells. VEGF-C mRNA and protein expression of all groups were compared. Results VEGF-C mRNA expression of negative control group and the control group showed no significant difference (P〉0.05). VEGF-C mRNA expressions of experimental groups were lower than negative control group and the control group (P〈0.01); expression of experimental group A was lower than expressions of experimental group B and C (P〈0.01). VEGF-C protein levels in culture super-natant of negative control group and the control group showed no significant difference(P〉0.05);VEGF-C protein levels in culture supernatant of expressions of experimental groups were lower than negative control group and the control group (P〈0.01); Level of experimental group A was lower than expressions of experimental group B and C (P〈0.01). Conclusion siRNA expression vector of targeting the VEGF-C can apparently decrease VEGF-C mRNA and protein expression on breast cancer cells.
