微波法提取金雀花多糖工艺研究 被引量:2

Study on Extraction Process of Polysaccharide in Parochetus communis by Microwave Method

摘要 采用微波法从金雀花中提取多糖,用苯酚-浓硫酸法测定其多糖含量。研究不同提取溶剂、提取方法对金雀花多糖提取率的影响,并通过单因素试验和正交试验对其多糖提取工艺参数进行了优化。结果表明,采用微波法提取金雀花多糖的最佳提取工艺参数为:微波功率800 W,微波时间2.0 min,料液比1∶40(g/m L)。在此条件下,金雀花多糖平均提取率为19.31%,加样平均回收率为98.92%,RSD值为0.23%(n=5)。该方法简便快速,准确度和精密度较高,回收率好,可作为金雀花多糖的提取和含量测定。 In this study, polysaccharide in Parochetus communis was extracted by microwave method, and the polysaccharide content was determined by phenol sulfuric acid method. Effect of different extraction solvents, exlraction method on the extraction rate ofpolysaccharide was also studied. The parameters of extraction process ofpolysaccharide were optimized through single factor experirnent and orthogonal experiment. The results showed that the optimum extraction parameters ofpolysaccharide by Microwave were as followed: microwave power 800 W, irradiation time 2.0 min, ratio of solid to liquid 1 : 40 (g/mL). Under such conditions, the average extraction rat of polysaccharide in Parochetus communis was 19.31%, the average recovery rate was 98.92%, RSD value was 0.23% (n=5). The method for extraction ofpolysaccharide in Parochetus communis has the advantages of simple operation, high accuracy, good reproducibility. Therefore it can be used in the extraction and determination of the content of the polysaccharide.
