参芪四逆汤对腹主动脉缩窄大鼠心肌纤维化的影响 被引量:3

摘要 目的:观察参芪四逆汤(党参、黄芪、附子、干姜、甘草)对腹主动脉缩窄大鼠心肌纤维化的影响,并探讨其对心肌转化生长因子β1(TGF-β1)和结缔组织生长因子(CTGF)的影响。方法:采用Doering腹主动脉缩窄法复制心肌纤维化模型。8周后,检测心脏血流动力学改变,Masson染色法观察心肌组织形态学改变,免疫组化法和western-blot法检测心肌TGF-β1和CTGF表达。结果:与模型组比较,参芪四逆汤可以明显改善大鼠心功能,减少胶原纤维含量,降低心肌TGF-β1和CTGF的表达。结论:参芪四逆汤具有防治心肌纤维化作用,其保护机制可能与其抑制心肌TGF-β1和CTGF表达有关。 Objective: To study the effect of Shenqi Sini decoction( Codonopsis pilosula,Astragalus mongholicus,radix aconiti carmichaeli, rhizoma zingiberis, liquorice )on myocardial fibrosis rats with abdominal aorta banding, and explore its effect on the expression of transforming growth factor β1 ( TGF-β1 ) and connective tissue growth factor (CTGF) in myocardium. Methods: The models were established by partially banding of abdominal aorta according to Doering' s method. Eight weeks later,hemodynamics changes of heart were tested,the changes of myocardial tissue morphology were observed by Masson staining, the expression of TGF-β1 and CTGF were tested by immunohistochemistry method and Western blotting. Results: Compared with the model group, Shenqi Sini Decoction can obviously improve heart function of rats,reduce the content of collagen fibers and depress the expression of myocardial TGF-β1 and CTGF. Conclusion: Shenqi Sini decoction can inhibit the expression of myocardial TGF-β1and CTGF, which may explain the mechanism of its protective effect on myocardium with fibrosis.
