不同培养条件对乳酸菌黏附猪小肠黏膜上皮细胞的影响 被引量:4

Effects of different culture conditions on Lactobacillus adhesion to the epithelial cells in porcine small intestinal mucosa

摘要 为了研究不同培养条件对乳酸菌黏附猪小肠黏膜上皮细胞的影响,试验采用体外细胞培养方法,观察乳酸杆菌在不同条件下(如pH值、温度、离子浓度)黏附猪小肠黏膜上皮细胞的能力。结果表明:p H值为6时,小肠黏膜上皮细胞黏附乳酸杆菌数为(15.60±0.58)个,较其他pH值时多;与其他温度(30℃和42℃)相比,37℃时猪小肠黏膜上皮细胞黏附乳酸杆菌数最多,为(22.44±1.05)个/细胞;与其他浓度相比较,猪小肠黏膜上皮细胞黏附乳酸杆菌数最多的是,2 mmol/L Ca^2+时为(11.36±2.36)个/细胞,2 mmol/L Mg^2+时为(18.75±1.37)个/细胞,0.005 mmol/L Fe^2+时为(22.48±2.78)个/细胞,0.000 5 mmol/L Cu^2+时为(20.24±2.35)个/细胞,0.0025 mmol/L Zn2+时为(17.48±1.59)个/细胞;不同浓度的S2-对乳酸杆菌黏附小肠黏膜上皮细胞黏附性影响不显著;0.005 mmol/L I-时小肠黏膜上皮细胞黏附细菌数为(19.68±1.25)个/细胞,其他浓度I-对乳酸杆菌的细胞黏附性影响不显著。说明乳酸杆菌与猪小肠黏膜上皮细胞黏附的最佳酸碱度、温度分别为pH值为6-7、37-42℃;Ca^2+、Mg^2+、Fe^2+、Cu^2+、Zn^2+、I^-等6种离子的最适浓度分别为2,2,0.005,0.000 5,0.002 5,0.005 mmol/L。 To study the effects of different culture conditions on Lactobacillus adhesion to the epithelial cells in porcine small intestinal mucosa, the method of cell culture, in v/tro was used to observe the ability of Lactobacillus adhesion to the epithelial cells in porcine small intestinal mucosa under different culture conditions, such as pH value, temperature and ion concentration. The results showed that the number of LactobaciUus adhesion to the cells were 15.60 ± 0.58 per cell with the pH value of 6 compared to the other pH values. The maximum number of Lactobacilli adhesion to the cells were 22.44 ± 1.05 per cell at the temperature of 37 ℃ compared With the other temperature (30 and 42 ℃ ). With the concentrations of 2 mmol/L Ca^2 + , 2 mmol/L Mg^2 + , 0. 005 mmol/L Fe^2 + , 0. 000 5 mmol/L Cu^2 + , and 0. 002 5 mmol/L Zn^2 + compared with the other concentrations, the maximum numbers of Lactobacillus adhesion to the cells were 11.36 ± 2.36,18.75 ± 1.37,22.48 ± 2.78, 20.24 ±2.35, 17.48 ± 1.59 per cell, respectively. The effects of different S2 - concentrations on the Lactobacillus adhesion to the epithelial cells in porcine small intestinal mueosa were not significant; the number of Lactobacillus adhesion to the cells was 19.68 ± 1.25 per cell with the concentrations of 0.005 mmol/L I ^-, but the effects of the other I - concentrations on the LactobaciUus adhesion to the cells were not significant. The results indicate that the optimal pH and temperature for LactobaciUus adhesion to the epithelial cells in porcine small intestinal mucosa were pH 6 to 7 and 37 ℃ to 42 ℃, respectively; and the optimum concentration of 6 kinds of ions including Ca^2 + , Mg^2 + , Fe^2 + , Cu^2 + , Zn^2 + , and I^ - were 2,2,0. 005,0. 000 5,0. 002 5,0. 005 mmol/L, respectively.
