摘要
斑马鱼(Danio rerio)一直作为模式生物研究脊椎动物的发育,在此过程中,斑马鱼的实验操作技术也得到了广泛发展。了解斑马鱼中枢神经系统(central nervous system,CNS)多巴胺能(dopaminergic,DA)神经元的定位及其投射对DA神经元功能的研究及利用斑马鱼建立人神经退行性疾病模型至关重要。此外,还能够更好地研究人神经退行性疾病的发病机制和探寻治疗方法。本文提供了很多证据证明斑马鱼CNS是构建人神经退行性疾病最适合的模型之一,还从发育学的角度阐释了DA神经元的发生及神经通路的形成在帕金森病(Parkinson's disease,PD)发病机制研究中的作用,并且对转基因斑马鱼模型在PD研究中的作用也做了较为详细的阐述。
As an animal model, zebrafish (Danio rerio) has been applied in exploring the development of vertebrates. Meanwhile,an extensive array of techniques for its experimental manipulation and analysis have been established. Understanding the location and projection of dopaminergic(DA) neurons in the central nervous system (CNS) of zebrafish would be vital to establish zebrafish as a human nerodegeneration model. In this review, we provide the latest evidence, which suggests that the CNS of zebrafish is an appropriate model for human neurodegenerative disease research. In addition,we review the role of DA neurogenesis and the formation of nervous pathway in Parkinson's disease (PD) ,from the perspective of development,and we also make a conclusion about the transgenic zebrafish model function as a propriate model in quest of mechanism of PD.
