Levels of IL-17 and IL-22 mRNA in the Blood of Type 2 Diabetic Nephropathy Patients

摘要 目的:研究2型糖尿病(T2DM)患者肾脏不同病变时期外周血 IL-17和 IL-22 mRNA 水平,探索糖尿病肾病(DN)患者 IL-17和 IL-22基因表达水平与肾脏病变的关系。方法实验包括60名2型 T2DM 患者和20名正常对照者(NC),其中糖尿病患者包括:无蛋白尿组(NA,n=22)、微量清蛋白尿组(MA,n=18)和糖尿病肾病组(DN,n=20)。采用实时荧光定量 PCR 方法检测 IL-17和 IL-22 mRNA 水平,分析各组外周血 IL-17和 IL-22 mRNA 水平差异。结果DN 组 IL-17和 IL-22 mRNA 水平显著高于 MA 组,NA 组和 NC 组差异有统计学意义(P <0.01);MA,NA 和 NC 三组之间 IL-17和IL-22 mRNA 差异无统计学意义(P >0.05)。结论在2型糖尿病肾病外周血中 IL-17和 IL-22 mRNA 水平升高,IL-17和 IL-22可能在 DN 病变中发挥作用。 Objective To investigate levels of IL-17 and IL-22 mRNA in the blood of type 2 diabetes(T2DM)patients with the different stages of kidney injury and explore the relationship between the gene expression levels of IL-17,IL-22 and renal lesions in patients with diabetic nephropathy(DM).Methods Subjects included 60 T2DM patients with or without kidney injury and 20 normal controls(NC,n=20).Diabetes patients were divided into 3 groups by level of urinary albumin to creatinine ratio (ACR):no proteinuria group (NA,ACR〈30 mg/g,n=22),microalb uminuria group (MA,30 mg/g〉ACR〉300 mg/g,n=18)and diabetic nephropathy group (DN,ACR〉300 mg/g,n=20).Quantitative Real-Time RT-PCR was used to detect IL-17 and IL-22 mRNA levels.Analysis differences of IL-17 and IL-22 mRNA levels among NA,MA,DN and NC groups.Results The levels of IL-17 and IL-22 mRNA were significantly higher in DN group than that in MA,NA and NC group (P 〈0.01,respectively),However,there were not significant difference among MA,NA and NC group (P 〉0.05,respectively).Conclusion Levels IL-17 and IL-22 mRNA were increased in blood of T2DM patients with nephropathy.IL-17 and IL-22 may play role in the pathogenicity of diabetic nephropathy.
