摘要
通过尼罗蓝平板筛选、苏丹黑染色法及摇瓶复筛,从土壤中分离筛选到一株高产PHB的菌株FQB6,经形态观察、生理生化实验及16S r DNA遗传分析,鉴定该菌株为蜡状芽孢杆菌(Bacillus cereus)。进一步研究菌株的生长规律,发现菌株的延迟期为0-36h,对数生长期为36h-60h,稳定期为60h-96h,当培养时间为84h时,PHB产量和生物量均达到最大,分别为1.926g/L和1.05g/L。
The strain FQB6 producing PHB isolated from soil by Nile blue plate, Sultan black staining and shake flask screening was identified as Bacillus cereus according to its morphological, physiological properties and 16S rDNA gene analysis. Growth regularity of further research on strain, strain found the lag phase, logarithmic growth and stationary phase was 0 -36h, 36h -60h and 60h -96h respectively when the incubation time for 84h, the yield of PHB and weight of dry cell reached the maximum, 1. 926g/L and 1.05g/L respectively.
