摘要
从人工老化后的不同发芽率水平的老芒麦干种子中提取基因组DNA,检测表明其可用于老芒麦种质资源的ISSR分析.ISSR标记分析表明,种子老化引起的老芒麦干种子基因组DNA分子片段的变化主要集中在750~2000bp和250~500bp范围,主要表现为DNA大分子片段的消失或颜色逐渐变浅,某些特异DNA片段出现,且其丢失和增加是随机的.
Dry seed genomic DNA was extracted after artificial aging to be used for ISSR-PCR genetic integrity analysis of Elyrnus sibiricus L. germplasm resources. The changes of dry seed genomic DNA fragments of Elymus sibiricus L. caused by seed aging mainly concentrated in 750--2000bp and 250-- 500bp and showed that macromolecular fragments of DNA disappeared and color became shallow gradually. Some specific DNA fragments appeared, and the lost and increase of genomic DNA fragments were random.
