高危型HPV感染与宫颈病变组织IFN-γ、IL-10表达的相关性研究 被引量:14

摘要 目的:研究高危型HPV感染与宫颈病变组织干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)表达的相关性。方法:将在本院就诊的HPV16感染患者、HPV18感染患者和非高危型HPV感染患者纳入研究,收集宫颈组织后分别采用荧光定量PCR和western-blot方法检测IFN-γ、IL-10、Toll-样受体4(TLR4)、依赖髓样分化因子88(MyD88)、NF-kB、ANT、PD-L2、Survivin含量。结果:(1)细胞因子:HPV16感染组、HPV18感染组的IFN-γ含量低于非高危型HPV感染组,IL-10含量高于非高危型HPV感染组,差异有统计学意义(P<0.05);(2)TLR4信号通路:HPV16感染组、HPV18感染组的TLR4、MyD88、NF-kB的含量均高于非高危型HPV感染组,差异有统计学意义(P<0.05);(3)凋亡调节分子:HPV16感染组、HPV18感染组的ANT含量低于非高危型HPV感染组,PD-L2、Survivin含量高于非高危型HPV感染组,差异有统计学意义(P<0.05)。结论:高危型HPV感染会抑制Th1细胞功能、增强Th2细胞功能、激活TLR4信号通路、诱导细胞增殖,与宫颈疾病的进展密切相关。 Objective.. To study the correlation between high-risk HPV infection and γ-interferon and interleukin-10 ex- pression in cervical lesions. Methods: Patients with HPV16 infection, HPV18 infection, and patients with non-high risk HPV infection that treated in our hospital were enrolled. Cervical tissues were collected and IFN-γ, IL 10, TLR4, MyD88, NF-kB, ANT, PD-L2, Survivin were detected by fluorescence quantitative PCR and Western blot. Results: (1) Cytokines : IFN-γ contents of HPV16 infection group and HPV18 infection group were lower than that of non-high risk HPV infection group; IL-10 contents were higher than that of non-high risk HPV infection group; (2) TLR4 signal pathway: TLR4, MyD88,NFkB con tents of HPV16 infection group and HPV18 infection group were higher than those of non high risk HPV infection group; (3) apoptosis regulatory molecules: ANT content of HPV16 infection group and HPV18 infection group were lower than that of non-high risk HPV infection group; PD-L2, Survivin contents were higher than those of non-high risk HPV infection group. Conclusion: High risk HPV infection can inhibit Thl cell function, enhance Th2 cell function, activate TLR4 signal pathway,induce cell proliferation; is's closely correlated with the progress of cervical disease.
