摘要
目的:优选出纯化胡芦巴提取物中4-羟基异亮氨酸的大孔树脂,并确定相应的最佳工艺条件。方法:以胡芦巴提取物中4-羟基异亮氨酸含量为评价指标,采用柱前衍生-高效液相色谱法检测24种型号大孔树脂静态吸附试验前后4-羟基异亮氨酸(4-HIL)含量变化来考察其纯化效果。结果:24种型号大孔树脂对4-HIL吸附容量均很低,但ADS-5型大孔树脂可用于胡芦巴提取物中4-HIL初步纯化,最佳工艺条件为采用ADS-5型大孔树脂,上样液浓度为0.04 g·m L-1,上样量为60 m L,上样流速为2 BV·h-1,收集续滤液蒸干可得。结论:ADS-5大孔树脂可用于其初步除杂。
To select the macroporous resins for the purification of 4-hydroxyisoleucine in the extraction of Trigonella foenum-graecum L., and to optimize the technical process, with the content of 4-hydroxyisoleucine as index, using pre-column derivatization of HPLC, the content of 4-hydroxyisoleucine before and after using 24 kinds of macroporotts resins was determined. The static saturated absorption capacity of 24 kinds of macroporous resins to 4-hydroxyisoleucine was very low, yet ADS-5 macroporous resins can be used for the preliminary purification of 4-HIL; the best effective purification was found in the experiment with ADS-5 macroporous resins with the concentration 0.04 g·mL^-1, sampling amount 60 mL, and the sampling rate 2 BV.h-1, collecting and drying the subsequent filtrate. ADS-5 macroporous resins can be used in preliminary removal of impurities.
