摘要
以21-脱氧可的松为原料,探索了一条合成醋酸可的松的新路线,并对路线中的脱保护反应进行了研究。通过对脱保护反应机理的探讨和正交实验找到了最优的反应条件,当T=25℃,t=3h,n(溴代盐)∶n(K2CO3)=1∶3,V(乙醇)∶V(水)=4∶1时,产品溴代物的收率最高,达到了84.57%,通过验证,收率稳定,适合工业化生产。
The new synthesis of cortisone acetate is starting from 21 - deoxyco - rtisone, and we do research to the deprotection in it. Through discuss the reaction mechanism of deprotection and orthogonal experiment, the best reaction cond - ition are got,when the reaction temperature is 25 ℃ ,reaction time is 3 hours ,molar ratio of bromine salt and potash is 1 : 3 ,volume ratio of ethanol and water is 4 : 1 ,the yiel - d of bromide is 84.57%. And the yield is stable through certification, so the process is suitable for industrial production.
