高血压病患者的PZ内含子FG79A基因多态性及蛋白Z水平实验观察

摘要 目的探讨本地区汉族人群中蛋白Z内含子FG79A基因多态性的发生率及血浆PZ水平与高血压病的相关性和临床意义。方法对高血压病组185例和对照组186例采用聚合酶链反应和限制性内切酶片段长度多态性方法并结合基因检测技术检测蛋白Z内含子FG79A基因多态性。同时检测两组PZ水平,观察其变化情况。结果发现本地区汉族人群存在蛋白Z内含子FG79A基因多态性,GG型、GA型和AA型分别占18.33%、51.75%和29.92%。G、A等位基因频率分别为44.20%和55.80%。高血压病组与对照组间基因型和等位基因频率分布差异无显著意义(均P>0.05);轻度、中度、重度高血压患者间基因型和等位基因频率分布差异亦无显著意义(均P>0.05)。高血压病组未发生心肌梗死和脑梗塞患者PZ水平为(1559.6±204.6)ug/L,发生心肌梗死和脑梗塞患者PZ水平为(801.0±201.0)ug/L和(650.0±376.2)ug/L,分别与对照组PZ水平为(1127.0±117.9)ug/L相比,F方差64.9,组间比较Q检验,均具有统计学意义(P<0.05)。结论皖南地区汉族人群存在蛋白Z内含子FG79A基因多态性,但此基因多态性与高血压病的发生发展无关联,高血压病不同组别血浆PZ水平的变化与自体凝血机制发生消耗有关。 Objective To investigate the distribution of protein Z intron FG79 A polymorphisms in Chinese and the associations with hypertensive disease and clinical significant. Methods The test group was consisted of 185 hypertension patients,the control group was consisted of 186 healthy people. The protein Z intron FG79 A polymorphisms were studied by PCR and restriction fragment length polymorphism. Obeserving two groups’ PZ changes by measuring PZ levels. Results The protein Z intron FG79 A polymorphisms were found in Chinese in Anhui province. The percentages of GG / GA / AA were 18. 33% /51. 75% /29. 92%. The frequencies of G and A alleles were 44. 20% and 55. 80%. There were no significant differences in protein Z intron FG79 A genotype and alleles frequencies distribution between test group and control( P 〉 0. 05). Neither significant differences were found among light,moderate and severe hypertension patients( P 〉 0. 05). The PZ of test group excluding miocardial infarction and cerebral infarction is( 1559. 6 ± 204. 6) ug / L. The PZ of miocardial infarction and cerebral infarction are( 801. 0 ± 201. 0) ug / L and( 650. 0 ± 376. 2) ug / L. The PZ of control group is( 1127. 0 ± 117. 9) ug / L. F variance 64. 9,group compareson Q test,there are significant difference between test and control groups( separately,P 〈 0. 05). Conclusion Protein Z intron FG79 A polymorphisms are exist in Chinese in south of Anhui province. Protein Z intron FG79 A polymorphisms were not associated with hypertensive disease,and the decreased PZ level in different patients with hypertension may be associated with a depletion of coagulation protein.
